| Objetives: This research is designed to clarify the function of Lian-yu capsule in lowering blood pressure and improvement of heart function; To provide scientific evidence for Lian Yu capsule in the treatment of hypertension and improving the function of the heart. The research also uses microholography technology to measure and calculate thickness of middle tunica and diameter of aorta and mesentery artery in SHR, in order to validate Lian-yu capsule have function on vascular remodeling of SHR.Methods:Pick up 24 SHR and 8 WKY that grow 13 weeks. All SHR are divided randomly in three groups:Model group, Valsartan group and Lian-yu capsule group, each group have 8 SHR, 8 WKY are the Normal group, 4 male and 4 female. Feed model group with physiological saline 0.9%, Valsartan group with liquor Valsartan and Lian-yu capsule group with Lian-yu capsule, 10ml/kg once a day last for four weeks. Measure all the rats'tail artery pressure before filling the stomach, 2h, 4h, 6h, 8h after it and from the first to the forth week. 4th weekend in rats under anesthesia for cardiac catheterization room measuring heart rate, and hemodynamic parameters of left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) Left ventricular pressure maximal rate (+dp/dtmax) Left ventricular pressure largest decline rate (-dp/atmax). Then cut right atria to form a channel to let the injection go through. Inject 300ml Paraformaldehyde 4%, after 30 minutes, open their thorax and celiac cavity, collect aorta 6mm, mesentery artery 4mm respectively, and put them in 20ml Paraformaldehyde 0.4% for 24 hours. Then dehydrated, clarified and darepose them with olefin. At last, make them 4μm aorta slice, and dye with HE. Observe the change e of aorta and mesentery arteryunder telescop, use microholography system to measure and calculate thickness of middle tunica and diameter of vessel cavity of SHR aorta and mesentery artery.Result:⑴The effect on blood pressure in SHR by Lian-yu capsule:The blood pressure of Model group SHR is significantly higher than Wky group. There was extremely significant difference (P <0.01). Compared with Model group, the blood pressure of SHR in Valsartan group and Lian-yu capsule group were significantly decreased (P <0.01) ⑵The effect on blood pressure in SHR immediate impact by Lian-yu capsule: Compared with Model group, the SHR in Lian-yu capsule group, in 2hours,4hours lower blood pressure have extremely significant difference(sP<0.01), and in 6 hours, 8 hours lower blood pressure was also a significant difference(P<0.05).⑶The effect on heart function in SHR by Lian-yu capsule: The LVEDP of Model rats increased, the LVSP and±dp/dtmax decreased. Compared with Wky group, there was extremely significant difference (P <0.01). Compared with Model group, the LVEDP in Valsartan group decreased and LVSP and﹣dp/dtmax increased, which had significant difference (P <0.05),﹢dp/dtmax increased, which had extremely significant difference (P <0.01); the LVEDP in Lian-yu capsule group decreased, which had extremely significant difference (P <0.01), LVSP,±dp/dtmax increased, which had significant difference (P <0.05)⑷Observe under telescope, WKY group, rats' aorta wall medium tunica and inner tunics are not thicken, vascular cavity are larger comparatively. Model group rats' aorta wall become thicker, especially the medium tunica, vascular cavity are smaller than WKY, medium tunica leiosporous cell are a little dareorder. Compared with SHR, Valsartan group and Lian-yu capsule group rats' aorta wall inner tunica and medium tunica become thinner, vascular cavity morbid symptom relieves in some degree. Compared with Wky group, Model group rats' aorta wall medium tunica/diameter of vascular cavity are more than that of Wky group, and they have extremely significant difference (P<0.01). Compared with Model group, Valsartan group and Lian-yu capsule group rats' aorta wall medium tunica/diameter of vascular cavity both reduce in some degree, and they have significant difference (P<0.05).⑸Wky group rats' mesentery artery medium tunica and inner tunica are not thicken, vascular cavity are larger comparatively. Model group rats' mesentery artery becomes thicker, especially the medium tunica, vascular cavity are smaller than Wky, leiosporous cell layer are increasing and dareordered. Compared with SHR, Valsartan group and Lian-yu capsule group rats' mesentery artery inner tunica and medium tunica become thinner, vascular cavity morbid symptom relieves in some degree.Compared with Wky group, Model group rats' mesentery artery wall medium tunica/diameter of vascular cavity are more than that of Wky group, and they have extremely significant difference (P<0.01). Compared with Model group, Valsartan group and Lian-yu capsule group rats' mesentery artery wall medium tunica/diameter of vascular cavity both reduce in some degree, and they have significant difference (P<0.05).Conclusion:1. Lian-yu capsule can reduce blood pressure in SHR and the effect is insistent and stable.2. Lian-yu capsule can improve the heart function of SHR.3. Lian-yu capsule can restrain vascular remodeling. |
