Primary liver cancer is a malignant tumor of the digestive system that can cause great harm to human life and health..Every year,the new cases of liver cancer worldwide and the deaths case of liver cancer are gradually increasing.Currently,the molecularly targeted drug sorafenib for clinical treatment of patients with advanced liver cancer is currently not stable or effective in delaying the survival of patients with advanced hepatocellular carcinoma,which may be different from those of patients with liver cancer.There is a certain relationship between the differences;however,there are literature reports showing that in the clinical use of sorafenib targeted therapy,combined with the use of treatment such as TACE treatment,local ablation therapy,the two treatment methods combined To make the two advantages can achieve complementary results.At present,it is a hot spot in the field of liver cancer research to study the clinical diagnosis and treatment of liver cancer,and find effective biomarkers to predict the prognosis of liver cancer.However,the study of the causes of liver cancer and the related molecular-level mechanisms in the occurrence and development of liver cancer are still very limited,and there is still no complete set of exact theories to explain this.Therefore,in-depth study of the relevant oncogenes in the progress of liver cancer in the process of the specific role played by what function and its molecular mechanism of the show what kind of mechanism to improve the clinical diagnosis of liver cancer and treatment levels and prognosis are evaluated of particular importance.Studies have shown that the STIL gene as a centriole replication-related factors in the formation of the centriole and the number of regulation,spindle formation and chromosome segregation plays an important role,which by affecting spindle formation and chromosome segregation,and thus involved in the occurrence and development of tumors,but its specific mechanism of action is not yet clearly stated.A number of studies have shown that STIL gene expression is elevated in many cancers and its high expression is related to tumor recurrence,invasion and prognosis,but the effect of STIL gene on hepatocellular carcinoma and its role in the development and progression of hepatocellular carcinoma mechanism of action,at present,the lack of relevant research reports.Objective To explore the expression of STIL gene in hepatocellular carcinoma and its prognostic significance.Methods The expression and prognostic value of STIL gene were initially predicted by Oncomine and TCGA.And then,67 cases of HCC patients tissue samples and matched normal tissues were retrospectively collected and analyzed by immunohistochemistry.STIL gene expression level was measured.Kaplan-Meier univariate and Cox multivariate analyses were used to explore the relationship and prognostic value between STIL expression and HCC patients.Results The Oncomine database predicted that the mRNA level of STIL gene was significantly higher in HCC than that of normal tissues(P <0.05).TCGA data showed that HCC patients with high mRNA expression of STIL gene had significantly lower overall survival than that of patients with STIL low expression(P=0.003).Subsequent immunohistochemical results confirmed the above predictive results,and the levels of STIL protein in HCC were highly expressed(P=0.001).HCC patients with high protein expression of STIL had dramatically decreased OS than that of patients with STIL protein low expression(P<0.001).Univariate and multivariate survival analyses revealed that STIL high expression could be used as an independent predictor of poor prognosis in HCC patients(All P values <0.001).Conclusion STIL gene expression in hepatocellular carcinoma increased,and high expression of STIL gene can be used as a prediction of poor prognosis of liver cancer patients were independent predictors.In the future,STIL gene is promising as a new target for the clinical treatment of liver cancer patients.
