Backgroud and Objective Acute lymphocytic leukemia(ALL)is a hematopoietic malignancy,originated from hematopoietic stem cells,which is characterized by primitive and immature lymphoblast cell clonal proliferating,suppressig narmal hematopoiesis,invading lymphatic tissue or extramedullary inflitration.Through standardized treament,complete remission rate of ALL patients could achieve 70% to 90%.Three years to five years disease free survival is 30% to 60%.Long non-coding RNAs(lnc RNAs)are a class of non-protein coding RNA transcripts longer than 200 nucleotides,which are accounted for total RNA 90%.The majority of lnc RNAs transcripted from RNA polymerase?have different shear variant.The expression of lnc RNAs have obvious tissue specificity and space-time specificity.Meanwhile,lnc RNAs have been implicated in diverse biological proccesses,such as cellular differentiation,proliferation,apoptosis and so on.Recently,emerging studies have show that aberrant lnc RNAs participate in the development of ALL and promoting leukemic cell chemotherapy resistance.Currently,the expression of lnc RNA RP11-87C12.5 in acute lymphocytic leukemia has not been reported.This study aim to investigate the expression of lnc RNA RP11-87C12.5 in acute lymphocytic leukemia and its clinical significance.Methods This study divide into three groups,that control group,de novo ALL patients group,and complete remission ALL patients after chemotherapy group.17 cases healthy people or nonmalignant hematopathy,served as control group,come from March 2016 to May 2017 in the first affiliated hospital of Zheng Zhou university in hospital or examine in outpatient service.De novo ALL group include contemporaneous 33 cases newly diagnosed ALL,which contained 19 cases of male,14 cases of female,10 cases of children and 23 cases of adult.In the 33 cases,there are T-ALL 6 cases,B-ALL 27 cases.The median age was 23 years old(one to sixty years old).There are 26 cases ALL after chemotherapy reache complete remission,that included 3 cases of pediatrics,23 cases of adult.Diagnosis and type according to WHO2016 criterion.Lnc RNA RP11-87C12.5 expression were evaluated in bone marrow samples from 17 control group,33 de novo ALL patients group,and 26 complete remission ALL patients after chemotherapy group by RT-PCR.At the same time the clinical data were collected and the clinical significance of lnc RNA RP11-87C12.5 expression was analyzed.Results 1.Compared with control group,lnc RNA RP11-87C12.5 is high expression in untreamt ALL group(P=0.021).Compared with de novo ALL group,lnc RNA RP11-87C12.5 is low expression in complete remission ALL group(P=0.039).2.In 33 de novo ALL patients,the expression of lnc RNA RP11-87C12.5 is no relation with patients' gender,age,T or B type,WBC,HB,PLT,LDH,proportion of bone marrow blast cell,BCR/ABL fusion gene,chromosome,WT1 gene,whether extramedullary infiltration,whether complete remission after one chemotherapy,whether relapse.3.In 27 B-ALL patients,compared with hight expression c CD79 a group,lnc RNA RP11-87C12.5 is hight expression in low expression c CD79a group(P=0.004).While it is no relevant with the other immunophenotype.Conclusion Lnc RNA RP11-87C12.5 is high expression in untreamt ALL group,low expression in complete remission ALL group.In B-ALL patients,compared with c CD79 a high expression group,lnc RNA RP11-87C12.5 is high expression in c CD79 a low expression group.It is indicated that lnc RNA RP11-87C12.5 may participate the development of ALL.
