Objective:To explore the different expression levels of three different transcripts of lncRNA RP11-191L9.4 between castration resistant prostate cancer cell lines PC-3 and DU-145.To explore the effect of RP11-191L9.4 on the proliferation,migration,apoptosis and tumor formation ability of cell lines PC-3 and DU-145.Method:qPCR assay was performed to explore the different expression levels of three different transcripts of RP11-191L9.4 between cell lines PC-3 and DU-145.To construct the virus vector that can knock down the expression of the transcript NO.1 of RP11-191L9.4.CCK-8 assay was used to observe on its role in cell proliferation.Transwell migration assay was used to observe on its role in cell migration.Flow cytometry(FCM)was used to observe on its role in cell apoptosis.Tumor formation assay was used to observe on its role in tumor formation ability.Results:The expression levels of three different transcripts of RP11-191 L9.4 in PC-3 cells is 53.87%、42.68%、3.45%.The expression levels of three different transcripts of RP11-191L9.4 in DU-145 cells is 75.62%、19.72%、4.66%.The expression level of transcript NO.1 of RP11-191 L9.4 is knocked down by the virus vector.The ability of cell proliferation can be significantly suppressed in CCK-8 assay.The ability of cell migration can be significantly suppressed in transwell migration assay.Cell apoptosis can be significantly promoted in flow cytometry(FCM).The ability of tumor formation can be significantly suppressed in tumor formation assay.Conclusion:The highest expression level of IncRNA RP11-191L9.4 in PC-3 and DU-145 cells is the transcript NO.l.Knockdown the transcript NO.1 of RP11-191L9.4 can significantly suppress the ability of cell proliferation,cell migration,promote cell apoptosis and suppress the ability of tumor formation.
