PART1 The study of changes of serum sex hormones and25(OH)-D3 levels in postmenopausal female BPPV patientsObjective: To explore the changes of serum sex hormones and 25(OH)-D3 level in postmenopausal female BPPV patients.Methods: We collect clinical data,including age,height,weight,medical history,physical examination and blood samples of the subjects,who satisfy the inclusion criteria and exclusion criteria in the vertigo center.Electrochemical luminescence was employed to detect the serum levels of E2,P,FSH,LH,25(OH)-D3.Results:We recruited 126 subjects(63 in the control and 63 in the experiment group)for our research from September 1st,2015 to February 1st,2017.There was no obvious difference in terms of age,BMI,hypertension,and diabetes in two groups.Compared with the control group,the serum levels of E2 and 25(OH)-D3 were significantly lower in the experiment group;and the serum levels of P,LH,FSH showed no significant difference in the experiment group.However,there was no relation between E2 and 25(OH)-D3 by correlation analysis.We also did correlation analysis between E2 and age.There was a lowly negative correlation between age and estradiol values in control group,correlation coefficient is-0.263.However,statistical significance was missed in the experiment groups.Conclusions:Compared with control group,the serum levels of E2 and 25(OH)-D3 are much lower in postmenopausal female patients.There was no relation between E2 and 25(OH)-D3 by linear regression analysis.Estradiol and 25(OH)-D3 deficiency are independent risk factors for BPPV in postmenopausal female patients.PART2 Regulation of otoconin90 by estradiol as a underlying mechanism of BPPV Objective: To study the regulation of otoconin90 by estradiol as a underlying mechanism of BPPV.Methods:Either a bilateral ovariectomy or a sham operation for the adult female rats was performed to establish the animal model.One month after the operation,either drugs or vehicle was injected at the nape of the neck for five group rats(including?OVX+Veh?OVX+P?OVX+E2?OVX+E2+P).Blood samples were collected from the tail vein of each rat before the operation,at the beginning and end of the therapy to determine serum E2 and P levels.Six hours after last injection,rats were sacrificed and inner ears were to detect the expression of OC90 protein and m RNA.The otoconia morphology of SHAM,OVX+Veh and OVX+E2 rats were observed by SEM and the expression of ER?,ER?,ERR? were also analyzed with WB.Results:The expression levels of OC90 protein,m RNA and ERR? were decreased and the otoconia were looser and have lower density after OVX.E2 replacement therapy,not P,was able to reverse these changes.ER??ER? and ERR? may involve in the regulation of otoconin90 by estradiol.Conclusions : Estradiol may regulate the expression of otoconin90,which is essential for the formation and development of otoconia,at the level of transcription through ER?,ER? and ERR?.
