Abstract/Summary:
Objective: To investigate the allele frequency distribution, gene diversity, geneticdistance and cluster analysis of17Y-specific short tandem repeats (STR) in TaiyuanHan population and evaluate their forensic application.Methods: We randomly selected121unrelated male individuals’ blood sample ofTaiyuan Han population. Their DNA extracted with chelex-100and detected17Y-STR loci with Y-filerTMsystem, and then used3130xl Genetic Analyze analysesthem. The allele frequencies and haplotype diversity were calculated. Y-STR data ofanother population were collected from publications, cluster analysis and phylogenictrees were applied to show the genetic distance among the populations.Results: The Y-filerTMsystems have137alleles. A total of120haplotypes wereidentified and the observed haplotypes diversity value was0.9999. The gene diversity(GD) values for each locus ranged from0.4180(DYS391) to0.8893(DYS385a/b).Compared with those Y-STR data of other populations were collected frompublications, the genetic distance between Taiyuan and Beijing Han populations wasthe smallest (0.0017), while the genetic distance between Taiyuan and Taiwan Atayalpopulation was the largest (0.4696). Compared with selected groups of minority, thegenetic distance between Taiyuan and Liaoning Manchu populations was the smallest(0.0014), while the genetic distance between Taiyuan and Taiwan Atayal populationwas the largest (0.4696).Conclusion: Multiplex detection of the17Y-STR loci revealed a highly polymorphicgenetic distribution, discrimination power (DP) and probability of exclusion (PE).They are very powerful for establishing a Y-STR database and forensic cases.
