Objective: 1.To analyze the caries levels in patients with different types of periodontal disease and whether there was any difference in periodontal health status between different caries susceptibility patients,and to explore whether there was related between dental caries and periodontal disease.2.The structure of salivary microbial community in aggressive periodontitis and caries-severe objects was studied by PCR-DGGE.The relationship between periodontal disease and dental caries was discussed from the perspective of microbiology,in order to provide new view of the pathogenesis of dental caries and periodontal disease.Methods: 1.According to different periodontal status,the patients were divided into four groups: the periodontal health group(HP group),the gingivitis group(GI group),the chronic periodontitis group(CP group)and the agressive periodontitis group(Ag P group).And according to different caries susceptibility status,the patients were divided into three groups: free caries group(FC group),moderate caries group(MC group)and severe caries group(SC group).The DMFT and CPI of the 229 patients from Lanzhou University School of Stomatology were analyzed by SPSS 22.0 statistical software.At the same time,compared the prevalence of periodontal disease in SC group.2.A total of 10 patients with aggressive periodontitis and 10 caries were enrolled as the research objects.Non-irritating saliva samples were collected and DNA was extracted.The 16 S r RNA gene was amplified and analyzed by DGGE.Diversity of community structure was analyzed using the software such as Quantity One 4.6.2 and SPSS 22.0 analyzedResult: 1.Compared with Ag P group,the DMFT of HP group,GI group and CP group were higher(P <0.05),which was statistically significant,indicating that low levels of caries in aggressive periodontitis.2.Compared with SC group,the CPI of FC group was higher(P < 0.05),which was statistically significant,suggesting that the periodontal condition of caries-severe patients was relatively health.3.Among the 136 patients in SC group,the detection rate of aggressive periodontitis was the lowest,only 2%;followed by chronic Periodontitis is 22%,suggesting that caries in patients with less simultaneous invasive periodontitis suggesting that cariessvere patients hardly suffer from aggressive periodontitis.4.Bacteria universal primers Bac1 and Bac2 were used to amplify the 16 S RNA gene of saliva samples.The amplified bands were detect by agarose and ultraviolet spectrophotometer,DNA main band is obvious and the concentration ?300 ng / ?l.5.16 cases of saliva samples PCR amplification products by DGGE,the map can be clearly seen.There is no exactly the same lane.The same experimental group include some common bright band.There were three bands in the caries group and two bands in aggressive periodontitis group.6.The similarity coefficient(Cs)was ranged from 0.3% and 43.4% in this two groups.The Cs of the aggressive periodontitis group and the caries group respectively were 24.6% ~ 76.3%,19.3 % ~ 73.4%.7.In the UPGAM phylogenetic tree,patients with caries and aggressive periodontitis develop two poly.The similarity coefficient was less than 0.21.The similarity of Ag-9 and Ag-13 in aggressive periodontitis group was 0.76.The similarity of CH-1 and CH-2,CH-5 and CH-3 in the caries group were 0.73 and 0.72,respectively.Conclusion: 1.The studies have consistently shown that caries and aggressive periodontitis are inversely related.2.There is a significant difference in salivary microbial community structure between patients with caries or aggressive periodontitis,indicating that salivary microbial community structure maybe play a role in the inverse relationship between the two diseases.
