Effects Of Shenwei Decotion On Chronic Kidney Disease With Kidney Deficiency Syndrome In Rats

Objective:To observe the changes of renal structure,function and modern medical indicators of kidney deficiency syndrome in rats with chronic kidney disease?CKD?after intervention of the Shenwei Decotion,so,we can investigate the therapeutic effect and mechanism of Shenwei Decotion on CKD with different kidney deficiency syndromes,further,to provide scientific basis for clinical application.Methods:1.150male SD rats were divided randomly into six groups,which were Nomal control group,CKD with yang-deficiency syndrome+normal saline group,CKD with yang-deficiency syndrome+Shenwei Decotion,CKD with ying-deficiency syndrome+normal saline group,CKD with ying-deficiency syndrome+Shenwei Decotion and CKD group.All group were intravenously injected with one-time adriamycin,and received adenine every two days by intragastric administration except the Nomal control group,2 weeks later,the rats in groups of A1-2?CKD with yang-deficiency syndrom+normal saline group,CKD with yang-deficiency syndrome+Shenwei Decotion?and B1-2?CKD with ying-deficiency syndrome+normal saline group,CKD with ying-deficiency synd rome+Shenwei Decotion?were separately received daily intragastric administration with thyroxine and pyrimidine for establishing CKD models with different kidney deficiency syndromes.We began to treat them for 4 weeks with Shenwei Decotion once the moldes successfully manufactured,the other two groups were treated with the same amount of normal saline,The above groups all were free to eating food,and drinking water.In the course of the experiment the general condition and the scores of kidney deficiency sydromes of therats were observed,testing the amount of urinary protein every week,in addition,the function of renal and Hypothalamus-pituitary-target glands were also assessed by the levels of the rats'serum creatinine,blood urea,thyroid hormone,thyroid stimulating hormone,testosterone,cyclic adenosine monophosphate/cyclic guanosinc monophosphate,and besides,HE and Masson staining were used to observe the changes of renal pathology in rats.and expression of TGF-?and IL-6 were measured by western blot.Results:1.After the models were established,the 24h urine protein,serum creatinine,blood urea nitrogen,as well as the level of renal fibrosis of all groups were significantly increased except the normal group?p<0.05?.2.Compared with the A1-2 groups and C group,B1-2 groups'scores of kidney deficiency-related behavior and appearance,cAMP/cGMP and the expression of lL-6 were all significantly increased?p<0.05?.3.After the rats in groups A₂ and B₂ were received drug intervention,the levels of24-hour urine protein,serum creatinine and blood urea nitrogen,as well as the degree of renal fibrosis all showed a significant decrease compared with the group of A1,B1 and C;the scores of kidney deficiency-related behavior and appearance,cAMP/cGMP and the expression of lL-6 were all close to the normal?p<0.05?.Conclusion:1.Based on the CKD Modes we could successfully use thyroxine and pyrimidine to create another innovative kidney deficiency syndrome models which more comfomed to the hunman'lesion of hypothalamus-pituitary-target glands.2.The Shenwei Decotion can significantly improved the structure and function of renal through down-regulating the expression of TGF-?,and inhibitting the activation of signaling pathways of TGF-?.3.The Shenwei Decotion can alleviate the kidney deficiency syndromes of CKD,the mechanism may be related to the regulation of cell function and systemicimmunity.
