Objective Cancer induces significant abnormalities in the blood system.Traditional medicine that the blood of the transport disorders in the occurrence and development of rock play an important role.This subject intends to analyze the hematological and pathological features of breast cancer mice and patients in different clinical stage.The abnormal hematopoietic progress of breast cancer was summarized.And an in vitro clone formation experiment of mouse spleen hematopoietic stem cells was used to explore the main differentiation-related factors of HSC in breast cancer,Looking for the relationship between blood transport disorders and the progress of breast cancer.MethodsSection One:We investigated the abnormalities of blood cell differentiation in breast cancer bearing mice by observing the lung metastasis,tumor volume and weight and peripheral blood parameters of breast cancer bearing mice.Section Two:Clinical data on breast cancer patients and healthy people were collected.The influences of lymph node metastasis,histological grade,BI-RADS classification,operation,radiotherapy and chemotherapy,on different peripheral blood parameters between patients and healthy people were analyzed.The relationship between neutrophil-lymphocyte ratio(NLR),platelet-lymphocyte ratio(PLR)and clinic pathological features of breast cancer was studied.The changes of blood parameters before and after therapies were compared.Section Three:The colon formation of the spleen cells in normal mice and tumor-bearing mice were respectively induced by normal serum,tumor-bearing mouse serum,Transforming growth factor-?(TGF-?)and cell culture medium.The numbers of Granulocyte macrophagic colony forming unit(CFU-GM),Erythrocytic burst-forming unit(BFU-E),Macrophagic colony forming unit(CFU-M)colons were counted.The percentages of different hematopoietic progenitor cells were analyzed by flow cytometry.ResultsSection One:Mice implanted with 4T1 breast cancer cells shown blood stasis and acute anemia.Blood test results showed a significant increase in white blood cell(WBC),number of neutrophils(NEUT#),number of basophils(BASO#)and number of eosinophils(EO#)and ratio of eosinophils(EO%)were significantly decreased,platelets(PLT)increased,plateletcrit(PCT)increased,laser doppler blood flow imaging(LDPI)showed decreased systemic perfusion.Section Two:1.Compared with the normal population,breast cancer patients with high ratio of neutrophil(NEUT%),NLR and PLR(P<0.05).Low ratio of lymphocyte(LYMPH%),EO%,LYMPH,EO,red blood cells(RBC),hemoglobin(HGB)and mean corpuscular hemoglobin concentration(MCHC)(P<0.05).2.After surgery,WBC,NEUT#,number of mononuclear(MONO#),RBC,HGB,hematocrit(HCT),platelet distribution width(PDW)were significantly decreased.EO%,mean platelet volume(MPV),PCT,PLR were significantly increased(P<0.05);After radiotherapy and chemotherapy,The patient's LYMPH,mean red blood cell volume(MCV),mean corpuscular hemoglobin(MCH),PLT,PCT were significantly decreased,monocyte ratio(MONO%),MONO,RBC,HCT and PDW were significantly increased(P<0.05).Futhermore,EO%,NEUT was significantly improved and close to normal levels,MONO,PLT were significantly improved after radiotherapy and chemotherapy and close to normal levels.3.Pearson analysis showed that,before surgery,the histological grade was negatively correlated with MCHC(r=-0.318,P<0.05),There was a significant positive correlation between the absolute value of LYMPH and the number of echo masses(r=0.296,P<0.05).MONO was negatively correlated with clinical stage(r=-0.356,P<0.05),and PLT was significantly positively correlated with the number of echo masses(r=0.320,P<0.05),BI-RADS grade was negatively correlated with PDW(r=-0.282,P<0.05),the maximum echo mass was negatively correlated with PDW(r=-0.491,P<0.01).After radiotherapy and chemotherapy,the Pearson correlation coefficient showed that,EO%was positively correlated with clinical stage(r=0.402,P<0.01).EO was associated with clinical stage(r=0.408,P<0.01).There was a positive correlation between EO%and BI-RADS grade(r=0.386,P<0.01).There was a positive correlation between EO and BI-RADS grade(r=0.374,P<0.01).4.Tumor with larger volume has higher NLR and PLR values,and the metastatic patients have higher PLR value than other patients,but not statistically significant(P>0.05).The relationship between NLR and tumor area after radiotherapy and chemotherapy was analyzed.The higher the area of tumor was found,the higher the NLR value was,the difference was statistically significant(P<0.01).Section Three:Clone formation assay indicated that tumor bearing mice has more CFU-GM,BFU-E,and CFU-M colon number(P<0.05).The effects of TGF-?(lng/ml)and tumor mice serum on clone formation were different.TGF-? could increase the number of CFU-GM colon and the percentage of CD115+ and Gr-1+/CD11b+progenitor cells.ConcjusionThe process of proliferation and metastasis of breast cancer induced abnormal phenomenon of the body with high white blood cells,neutrophils,platelet count,PLR and NLR,but low red blood cell count.The body showed chronic inflammation,blood stasis and anemia.The increase of peripheral blood immune negative regulation factor contributed to breast cancer escape.Tumor volume decreased or disappeared after surgery,radiotherapy and chemotherapy made the abnormal trend of normalization.The abnormal hematopoiesis in breast cancer bearing tumor may be related to the imbalance differentiation of hematopoietic stem cells induced by various growth factors such as TGF-P secreted by tumor.TGF-? can obviously promote proliferation of spleen hematopoietic stem cell in tumor bearing mice,promote the formation of CFU-GM colony in mouse spleen cells,and the differentiation of hematopoietic stem cells into megakaryocytes and myeloid progenitor cell differentiation were also increased.However,TGF-? has an inverse effect in spleen cells isolated from normal mice on the differentiation of hematopoietic stem cells.
