Abstract/Summary:
ObjectiveVerify the participation of bone marrow mesenchymal stem cell (MSC) in damage repair withA-549 lung cancer-bearing mice model; explore the mechanism of bone marrow mesenchymalstem cells participating in the tissue repair of tumor.MethodsIdentify hMSC isolated and cultured in vitro with flow cytometry and build A-549 lungcancer-bearing mice model. With peritumoral injection of hMSCs in experimental group,comparatively observe living condition of animal and size of tumor growth. Draw materials after4 weeks, comparatively observe tumor tissue with HE staining and compare collagenous fibrewith Masson staining. Detect the expression of FSP and FAP in two groups usingimmunohistochemistry to reflect the degree of interstitial fibers in tumor tissue in two groups.Compare the expression level of VEGF, HGF, IL-6 and Tn-C in two groups usingimmunohistochemistry.ResultsMSCs promoted tumor growth in tumor-bearing mice. There was no significant difference inliving condition of animals in two groups, the growth rate of tumor tissue in experimental groupwas faster than the control group, and the difference between groups was significant. Detect theexpression of a-SMA using RT-PCR, the expression of a-SMA mRNA was significantlyincreased in experimental group compared with control group. Using immunohistochemistry todetect the expression of tumor angiogenesis factors (TAFs) markers (FSP and FAP) in tumortissue of experimental group. The expression level of VEGF、HGF、IL-6 and Tn-C detected byIHC was significant higher than control group and the difference was statistically significant(P<0.05)..
