Comparative Study Of Pharmacodynamics Between The Traditional Zuojinwan And Ultra-Zuojinwan

Objective To study the effect of different doses of the TZJW and UZJW on rat's chronic gastritis and human gastric cancer cell SGC-7901,providing the scientific basis for the clinical application of Ultramicro Chinese medicine.Methods The effects of the TZJW and UZJW on chronic gastritis were compared by gastric acid,pepsin activity,SOD activity,MDA value and pathological section.CCK-8 method was used to determine the inhibitory effect of different dosage of TZJW and UZJW on the growth of human gastric cancer cell SGC-7901 at different time(24,48,72h).Hoechst staining was used to observe the morphological changes of human gastric cancer SGC-7901 cells treated with different doses of the TZJW and UZJW.Western-blot was used to detect the effect of different dosage of the TZJW and UZJW on the expression of Bax and Bcl-2 protein in human gastric cancer cell SGC-7901.Results In vivo experiment show that The UZJW and TZJW can reduce the pH of gastric and pepsin activity,increase serum SOD activity,reduce MDA content,while the results from the pathological section showed restoration of gastric mucosa.In vitro experiment show that different doses of UZJW and TZJW can significantly inhibit the activity of human gastric cancer cell SGC-7901 cells in a dose-dependent manner.Hoechst test results showed that different doses of the TZJW and UZJW can induce apoptosis on gastric cancer cell SGC-7901.It was found that different doses of UZJW and TZJW could significantly up-regulate the expression of pro-apoptotic protein Bax and down-regulate the expression of Bcl-2.In vivo and vitro experiments showed that the effect of UZJW is better than TZJW.Conclusion The same concentration of the TZJW and UZJW work as the same time,the effect of UZJW is better than the TZJW.
