Lipoic acid compound belonging to the vitamin B class,acts as a coenzyme in a multi-enzyme complex such as pyruvate dehydrogenase,alpha-ketoglutarate dehydrogenase,aminocaproic acid decarboxylase etc,Lipoic acid is the strongest known natural antioxidant.Glucose toxicity induced cells injury and apoptosis is an important aspect of the study of diabetes mellitus,but the super antioxidant effect of lipoic acid provides a new possibilities for the treatment of the cells injury caused by glucose toxicity.Germany had used lipoic acid treat diabetic neuropathy for more than ten years,the effect of lipoic acid in the clinical trials of diabetes is obvious.Amphibians dwelling in the midst of the wet conditions,the bare skin contains a variety of chemicals in order to adapt to the living environment and regulate physiological functions.Therefore,amphibians is the ideal model of the research of the species and structural characteristics of insulinotropic peptide.At present,the kinds and quantities of insulinotropic peptide that the international community had isolated from amphibians animals is the most.Some insulinotropic peptide has antibacterial effect.China is vast,the kind of frogs is variety,it is provide a favorable source of antimicrobial peptide and insulinotropic peptide isolated from frog skin.Antimicrobial peptide could improve a variety of drawbacks due to the traditional method of treating diabetes,and it makes antimicrobial peptide become the new main direction of improving insulin secretion and the treatment of type 2 diabetes.Antibacterial peptide Dybowskin-2CDYa(SAVGRHGRRFGLRKHRKH)is the new type antibacterial peptide which is found and named by our laboratory.AWRK6(SWVGKHGKKFGLKKHKKH)is the transformed peptide of our laboratory.The preliminary experiments of our Laboratory shown that the peptide has a well antibacterial activity and a variety of features of the frog skin insulinotropic peptide,can promote insulin secretion in pancreatic ? cells.In this paper,we used the lipoic acid injection produced by Shanghai Modern Hasson(Shangqiu Pharmaceutical Co.Ltd.)and the post-production synthetic of antimicrobial peptide Dybowskin-2CDYa AWRK6 as experimental raw materials,mouse pancreatic beta cell line MIN6 as experimental subjects,enzyme-linked immunosorbent assay ELISA and real-time quantitative reverse transcription polymerase chain reaction Real-time RT-PCR for the research methods research the protection of glucose toxicity of mouse pancreatic beta cell line MIN6 by add lipoic acid injection and antimicrobial peptide Dybowskin-2CDYa,AWRK6.Elaborate expound the recovery effect of the insulin secretion of pancreatic beta-cell and the reduced effect of the damage pancreatic beta-cell which is caused by glucose toxicity,it is expounded from protein and gene level by using lipoic acid injection and antimicrobial peptide Dybowskin-2CDYa AWRK6.The experimental results show that added lipoic acid injection,the quantity of insulin that damaged cells secreted can increased 0.54-1.27ng/ml,gene amplification increased 0.44-3.29ct.added antimicrobial peptide,the quantity of insulin that the damaged cells secreted can increased 0.68-1.57ng/ml,gene amplification increased 0.3-3.17ct.Annexin V-FITC flow cytometry and TUNEL in situ apoptosis detection show that the lipoic acid and antimicrobial peptide Dybowskin-2CDYa AWRK6 can reduce the apoptosis due to glucose toxicity,enhanced cell viability,restore cell function.
