Objective: By building type 2 diabetes mellitus (T2DM) rats model, this research aims to explore to observe the influence on the expression of GLUT4 of the muscle and the influence of MCP interentions. Methods: after one week of acclimation, 10 of the 40 six-week-old SPF male SD rats were randomly selected as normal group (NG) and given basic feed , other 30 rats were fed with hypercholesterol foods for one month and then randomized into three groups and intraperitoneally injected with small dosage of STZ: Group A: 30mg/kg (10 rats), Group B: 40mg/kg (10 rats), Group C: 50mg/kg (10 rats). Observe T2DM model established the success and approp riate STZ dose. Another 40 six-week-old SPF male SD rats were acclimated for a week and 10 of them were randomly selected as NG and given basic feed. The rest 30 SD rats according to the above method to to build T2DM SD rats model, Theses 30 rats were then randomized into three groups: T2DM Group (DM), MCP low dose group (LM) and MCP high dose group (HM). While continued to be fed with hypercholesterol foods, LM and HM were intragast -rically administered different dosage (1.5g/kg?d,3.0g/kg?d) of MCP for one month; DM were lavaged with distilled water. No food but only water was given for 12 hours. were anesthetized with an intraperitoneal injection of 10% Chloral Hydrate 350 - 400mg/kg, all were executed by the abdominal aorta bloodletting, and gather right hind leg thigh muscle tissue, Pathological changes were observed by HE staining. Immunohistochemical method was used to detect the expression of GLUT4 protein in of skeletal muscle tissue. Results: Group B: SD rats injected with STZ 40mg/kg show the best blood glucose stability and highest success rate and survival rate. After three weeks of MCP intervention, HM and LM show a lower blood glucose level compared with DM group (P<0.05).. MCP intervention group, atrophy is not appeared, muscle fiber bundle has not seen relatively narrow, neatly arranged obviously, the nucleus increased, position normal appearance roughly muscle horizontal grain compared with N group muscle. LM and no obvious difference HM intervention group. IHC: Compared with N group, the positive express quantity of GLUT4 of skeletal muscle of T2DM rats(including DM, LM, HM group), are obviously reduce (PDM < 0.01, PLM < 0.01, PHM < 0.05).Compared with DM, the positive express GLUT4 of skeletal muscle of HM rats quantity increased significantly (P < 0.05), and LM group, was not statistically significant (P > 0.05). Between different doses MCP intervention -n , positive express quantity of GLUT4 of skeletal muscle whose difference was statistically significant compared between LM and HM rats (P > 0.05). Conclusion: 1. Hypercholesterol diet and intraperitoneal injection of small dosage of STZ can successfully build Type 2 Diabetes Mellitus rats model. Group B: SD rats injected with 40mg/kg of STZ is the appropriate dosage. 2.MCP can reduce the blood glucose levels of T2DM rats, to a certain extent, improved DM group, necrotic and skeletal muscle fiber denaturation become pyknotic nuclei such pathological changes. 3. In a certain degree , skeletal muscle the decrease of protein expression GLUT4 of rat skeletal muscle is one of the pathogenesis of T2DM, and MCP rats in recovery under protein expression of GLUT4 of skeletal muscle of T2DM, and a certain dose dependent, tip high doses MCP through certain extent possible recovery under protein expression of GLUT4 of skeletal muscle of T2DM for clinical improvement IR, the prevention and treatment of T2DM provides new ideas and new choices.
