| ã€Objective】In this study, we want to observe the effects of DJB on glucose metabolism in GK rats and the effects of DJB + hepatic branch of vagus resection for non-obese type 2 diabetes mellitus(T2DM) rats glucose metabolism.And explore the hepatic branch of vagus in the regulation of glucose metabolism function. ã€Methods】Male Goto-Kakizaki(GK) 32 rats, 8-10 weeks, bodyweight 220-280 g,feeding in SPF conditions(provide from cavensla company in Changzhou). Then randomly divided into four groups: DJB group(retain full stomach,idle the duodenum, distal jejunum anastomosis with pylorus),DJB+hepatic branch of vagus resection group,sham operation group(cut off the pylorus part and resuture,make a 5cm longitudinal incision from the Treitz in 10 cm and resuture) and sham operation+hepatic branch of vagus resection group. Record the success rate of model and the survival rate of 1 week after operation.The changes of preoperative and postoperative 1,4,8,12 weeks body weight, fasting blood glucose,fasting serum GLP-1ã€PYY and FINS levels among four groups were observed. Statistical analysis these indexes. ã€Results】1.operation success and survival rate of animal1 rat dead of intestinal obstructionin the fourth day after the operation in the SO group(n=7). 1 rat excluded because of do not exactly cut off the hepatic branch of vagus in the SO+HBVR group(n=7).In the DJB group,2 rats dead of abdominal bleeding and anastomotic leakage in the 3,6day after the operation respectively(n=6). In the DJB+HBVR group, 1 rat excluded because of do not exactly cut off the hepatic branch of vagus and 1 dead of anastomotic leakage in the 2 day after the operation(n=6).The remaining rats completed the follow-up experiment.2. Body weightBefore operation, 4 groups of rats with body mass of at the same baseline. Compared with preoperation,the body weight of SO group and SO+HBVR group were decreased,but do not significantly.After the operation,the body weight of DJB group and DJB+HBVR group were significantly decreased compare to the peroperation(P<0.05).From the 4 week after operation, the DJB+HBVR group’s body weight was gradually increased,and the DJB group always decreased.3. Fasting blood glucose(FPG)Before operation, 4 groups of rats with the same fasting blood glucose. Compared with preoperation,the FPG of SO and SO+HBVR group have no differences. After the operation,the FPG of DJB group and DJB+HBVR group were significantly decreased compare to the peroperation(P<0.05);and compare to the SO group at the same time,both of the groups were significantly decline(P<0.05).4. Fasting serum PYYIn the reoperative there were no statistical significance of PYY content in these groups. Compared with preoperation, the fasting serum PYY of SO group have no differences. After the operation, Compare to the SO group,the SO+HBVR group have the lower PYY content in the same time. After the operation,the fasting serum PYY of DJB group and DJB+HBVR group were significantly increased compare to the peroperation(P<0.05).And compare to the SO group at the same time,both of the groups were significantly increased(P<0.05).In the same time point after the operation, DJB+HBVR group had the lower PYY content compare to the DJB group,and in the 8,12 weeks were significantly(P<0.05). 5. Fasting serum GLP-1In the reoperative there were no statistical significance of PYY content in these groups. Compared with preoperation, the fasting serum GLP-1 of SO group have no differences. After the operation, Compare to the SO group,the SO+HBVR group have the lower GLP-1 content in the same time,and at the 4 week after the operation were significantly decreased(P<0.05). After the operation,the fasting serum GLP-1 of DJB group and DJB+HBVR group were significantly increased compare to the peroperation(P<0.05).At the 8,12 weeks after the operation,the DJB group had significantly increased than DJB+HBVR group(P<0.05).6. Fasting Insulin Serum(FINS)In the reoperative there were no statistical significance of FINS content in these groups. Compared with preoperation, the fasting serum FINS of SO and SO+HBVR group have no differences. At 1week after the operation,the fasting serum FINS of DJB group and DJB+HBVR group were significantly increased compare to the peroperation(P<0.05). And compare to the SO group at the same time,both of the groups were significantly increased(P<0.05). ã€Conclusions】1. DJB+ hepatic branch of vagus resection is a feasible model;2. DJB can significantly reduce the body weight, fasting blood glucose innon-obese type 2 diabetic rats.At the same time, increase the serumGLP-1, PYY content and fasting insulin levels, it’s significantly improvethe diabetic state effect.3. Retain hepatic branch of vagus can be more durable to maintain low bodymass,and increasing the content of insulin, GLP-1, PYY. It played a role in theregulation of glucose metabolism. |
