Human development of the marine resources have become increasingly frequent,coming with the marine environmental pollution problem subsequently.Marine pollutants are include of heavy metals,pesticides,organic pesticide and others.The pesticides is one of the important pollutants of marine because of it has the characteristics of enrichment,refractory,high residual,and can enter into the Ocean with via runoff and rain erosion.Most of the pesticide is the environmental estrogen.The effects of pesticides for endocrine is physiologic function of mimic estrogen in the body.Kelp and Sargassum fusiformea is play an important role in the production and human life.Because it’s the function of balanced nutrition,good taste and health care,the consumption of it is increasing.At present,there are lots of researches on environmental hormone pollutants in seawater,sediment and marine animal(fish,shellfish,shrimp),but less research on Kelp and Sargassum fusiformea.This paper selects the Kelp and Sargassum fusiformea as the research object,analysis the estrogen activity by the residues pesticide.Breast cancer MCF-7 cells are classic cell lines with estrogen-sensitive characteristics;Sertoli cells provide nutrition for spermatogenic cells,and with the function of secrete a variety ofbioactive protein,horm one conversion,isolation barrier,phagocytic.Sertoli cells as a mature ovarian follicle granulosa cells in the largest number of the most versatile cells;granular cell provides a nutritional support for oocyte,also has important role of function and development for ovarian,granulosa cells of any damage substances will produce female reproductive toxicity.So this experiment selects the breast cancer MCF-7 cells,supporting cells,granulosa cell as the research object,compared the toxicity of pesticide residues to cells between the Kelp and Sargassum fusiformea.The experiment mixed with collagenase and trypsin digestion enzyme isolated from mouse Sertoli cells,ion chelator ethylenediamine diacetic acid(EDTA)and trypsin digestion enzyme extract mixed ovarian granulosa cells;Isolated cells and then using the resulting identification inverted microscope,HE staining,protein immunochemical method;By comparing the MTT assay seaweed,Sargassum pesticide residues on MCF-7 cells,support cells,affecting the proliferation of granulosa cells;Using Western blot assay extracts of pesticide residues in MCF-7 cells,support cells,the granulosa cells the MAPK signal transduction pathway related proteins ERK1/2,p-ERK1/2 protein expression distribution.Extraction of Sertoli cells,ovarian granulosa cells were identified by inverted microscopy,HE staining,protein immunochemical found cell purity of 80%.MTT results showed seaweed,Sargassum Pesticide Residues drug extract on MCF-7 cells,support cells,granulosa cell proliferation both,48h proliferation maximum effect with the negative control group had differences(P<0.05).Western Blot assay showed seaweed,Sargassum extract pesticide effects on MCF-7 cells,Sertoli cells,ovarian granulosa cells after 48h,positive drug estradiol(E2),the extract can significantly promote Sertoli intracellular ERK1/2,p-ERK 1/2 protein expression,with the negative control group had differences(P<0.05).So,seaweed,Sargassum extract pesticide residues may interfere with the estrogen activity.
