| Objective:Set up the rat models of kaolin PICA,discuss the curative effect of Wen Dantang to treat the Delayed vomiting induced by chemotherapy,and research it's mechanism of action.Methods:The first adaptive feeding rats,which means that quantify the normal feed and kaolin feed,and give the rats,and every time take the two feed out to observe its surface with or without bite marks and weighing,until the rats no longer eat kaolin.Then take 24 rats randomly divided into blank control group and cis-Dichlorodiamineplatinum in low,medium and high dose groups,and observe about the rats kaolin behavior after administration of 48h,record the the body weight of rats,the application amount of the normal feed and kaolin,and set up the rat models of kaolin PICA.Another 30 rats were randomly divided into blank control group,ondansetron group,Wen Dantang in low,medium and high dose groups after intragastric administration,after 30min 5 groups were intraperitoneal injection the optimal dose of cis-Dichlorodiamineplatinum,observe and record the The body weight of rats,the normal feeding cis-Dichlorodiamineplatinum and the Consumption of kaolin in 48h of rat after administration.Extract the Venous blood of rats after administration of 48h to determine the Vasoactine Intrestinal Peptide(VIP),Epidermal Growth Factor(EGF)and gastrin(Gas)in peripheral blood.After the rats In each group were killed by cervical dislocation,rapidly take out of the antral gastric tissue and slice it up for immunohistochemistry,observation of substance P expression.Results:1.Based on the successful establishment of rat model of kaolin,this experiment,through the determination of the body weight of rats,the consumption of ordinary feed and kaolin after the administration of physiological saline,ondansetron,Wen Dantang in small,Medium and high dose,showed that each group the body weight of rats decreased after administration of 24h,48h,and the statistical analysis showed that there were no significant differences in them(P>0.05);the average feed consumption decreased,which was more obvious in the Wen Dantang in small,Medium and high dose,but the statistical analysis showed that there were no significant differences in them(P>0.05);Rat kaolin phenomenon was more obvious,and the Rat kaolin consumption in ondansetron,Wen Dantang in small,Medium and high dose was less than blank control group in 24h,which has the Statistical significance(P<0.05),while there was little difference on the rats in 48h,and there were no significant differences in them(P>0.05).2.About the content of the VIP,EGF,GAS in peripheral blood of rat after treatment,the results showed that Wendan Decoction of low,medium and high dose group,ondansetron group of peripheral blood VIP,EGF,Gas levels were significantly higher than the control group,while the Wendan Decoction low,middle and high dose group,thecontent of different degree were higher than those ofondansetron group.Statistical analysis showed that the contents of VIP in peripheral blood of high dose group of Wen Dantang was significantly higher than that in the control group,and there was statistical significance(P<0.05);The contents of EGF in peripheral blood of Wen Dantang in medium,high dose group were all higher than that of control group and ondansetron group,and there was statistical significance(P<0.05);The contents of Gas in peripheral blood of Wen Dantang in high dose group were higher than that of control group and ondansetron group,and there was statistical significance(P<0.05)3.The Positive area,the total optical density of the blank control group,ondansetron group were slightly higher than that ofWendan Decoction of low,middle and high dose group,the average optical density of each basic almost.Statistical analysis showed that the positive area,average optical density and the total density of the gastric antrum,showed that there were no significant differences between each dose of Wen Dantang,ondansetron and the blank control group after administration(P>0.05),and there were also no significant differences between Each dose of Wen Dantang and ondansetron(P>0.05).Conclusion:1.The Wen Dantang has a certain effect to the delayed vomiting induced by chemotherapy,and there are differences while it was compared with the control group(P<0.05).2.The mechanism of action of Wen Dantang may be related with the increase of content of the VIP,EGF,GAS in peripheral blood,to relax the esophageal sphincter,increased mucosal blood flow and other relevant,and its mechanism is worthy of further investigation. |
