Objective:To uncover the anti-gastric ulcer effect in rats of the fibrous root part extract and the pseudobulb part extract of Bletilla striata with the modem pharmacology method.At the same time,the material base and mechanism of pharmacological activity was preliminary studied,and it will pave the way for the fibrous root resource utilization and new medicine exploitation.Methods:(1)Enrich the small organic chemical substances in the fibrous root part and the pseudobulb part of Bletilla striata with polyamide and analyze with HPLC method;Extract the polysaccharide of the pseudobulb part of Bletilla striata with water extraction and ethanol precipitation;Phenol-sulfuric acid method,3,5-dinitrosalicylic acid(DNS)method and Coomassie brilliant blue method were earried out to evaluate the polysaccharide of the pseudobulb part of Bletilla striata;(2)Gastric ulcer model of absolute ethyl alcohol to the SD rats was established,and the therapeutic effect of different extracts were evaluated through calculating bleeding area of the gastric ulcer which has hematoxylin-eosin staining with IPP 6.0;The SOD and MDA level in serum were detected,and the cytokines IL-lp,IL-10,PAF,VEGF and TNF-a level in serum were analyzed by ELISA;moreover,the expression level of IL-1?,IL-10,PAF,VEGF and TNF-a in gastric were detected by histological immunity method as well.Results:(1)HPLC indicated that the small organic chemical substances in the fibrous root part and the pseudobulb part of Bletilla striata were similar,but the content of the former was much higher than the later.The polysaccharide extracted from the pseudobulb part was mainly composed of high molecular saccharide,and without any protein.The content of total polysaccharide is 103.64%.The content of reducing sugar is 16.29%.(2)Anti-gastric ulcer research indicated that treatment with polysaccharide of pseudobulb part and small organic chemical substance of fibrous root part can prevent gastric damage effectively.61.08%bleeding area was inhibited treated by 200mg/kg small organic chemical substance of fibrous root part;however it was less effect than polysaccharide,which 71.91%bleeding area was inhibited when treated by 50mg/kg polysaccharide.HE staining revealed both treatment groups could prevent gastric mucosa damage and relieve gastric mucous hyperemia.Immune chemical and ELISA results indicated that,both polysaccharide and small organic chemical substances could prevent gastric damage by down regulating IL-1?,TNF-?,PAF and up regulating IL-10 and VEGF.Additionally,change of SOD and MDA level in serum revealed that both treatments could educe anti-gastric ulcer effect by up regulate SOD and down regulate MDA level.Conclusion:The small organic chemical substances in fibrous root part and pseudobulb part of Bletilla striata were consistent,with further research,the fibrous root part may become new medicinal part.And they all could prevent gastric damage in rats observably,mechanisms underlying it may be related to reduce the expression of inflammatory cell factor and rise the expression of inhibit inflammation response cell factor and improve oxygen radical scavenging activity of antioxidant system in the body.
