Objective:The present research aimed to investigate the anticancer activity of Radix Ranunculus Ternati abstracton Non-Small Cell Lung Cancer(NSCLC)and explore the possible mechanism.Then offer the thesis and experiment proofs for curing NSCLC by the extracts of Radix Ranunculus Ternati.Methods:?Radix Ranunculus Ternati Saponins(RRTS)and polysaccharides were extracted by the technique of solvent-refining.?3H-TdR incorporation assay was applied to detecting the inhibitive effect of different concentrations of RRTS on the proliferation of NSCLC cells,and calculate the median inhibitory concentration(IC50 value).? The cell cycle phase and apoptosis were inspected by flow cytometry.? Study the anticancer action of RRTS in human NSCLC A549 on nude mice.? The protein expression profiles of EGFR and MMP-9 were determined by immunity histochemistry technology.Result:?Radix Ranunculus Ternati Saponins(RRTS)and polysaccharides were extracted by the technique of solvent-refining,test results show that the content of RRTS and polysaccharides were 52%and 85%.?In vitro RRTS inhibited the growth of cell A549 and NCI-H460,showing a dose-effect curve,and the IC50 values were 44.3±11.2?g/ml,120.84±20.6?g/ml respectively,however the inhibition effect of polysaccharides was not obvious.The results indicated that RRTS is the antitumor effective components.?It found in FCM analysis that RRTS could promote the early apoptosis of cell A549 and NCI-H460,and arrest the cell cycle in G0/G1 phase.? In A549 xenografts model,the mice were sacrificed after RRTS administration for 18 days.The tumor inhibition rate(%)of RRTS 1.73g/kg group was 32.6,respectively(P<0.05).The results show that in vivo the RRTS abstract can obviously inhibit proliferation in human NSCLC A549 on cancer vaccinal nude mice..?The positive expression on percentage of EFGR and MMP-9 protein is lower in different concentrations of RRTS groups than the control group,showing a dose-effect curveand there was significant difference in the positive expression percentage of EFGR between the high RRTS and the control groups(P<0.05).Conclusion:This experiment preliminary confirm that RRTS could remarkable inhibit the growth of NSCLC cell and it was highly effective against nude mice transplanted tumor in vivo,and its mechanism was mailly about apoptosis induction and blockade of cell cycle in G0/G1 phase,down regulation of the protein expression level of EGFR and furtherinhibiting cell proliferation,down regulation of the protein expression level of MMP-9 and further the suppression of the tumor invasion metastasis et al.
