Objective:Recognize pathomechanism of Steroid-induced avascular necrosis of femoral head further by histopathological study on different joints or position in rats.And then to take Proteomics technology,find differential expression on protein,try to investigate the effect target and pharmacodynamic of Youguiyin treatment of SANFH.Methods:30 Wistar rats were involved,and were randomly divided into control group,experimental group and the Youguiyin group.SANFH models were established by injection of colibacillus endotoxin and prednisolone intramuscularly.Youguiyin group were lavaged with You Gui Yin,while control group and experimental group were lavaged with the same amount of saline.The behavior experiment lasted six weeks.6 weeks later,one side of femoral head,caput humeri,elbow joint and knee joint were used to Pathologic investigation.Bone tissue protein extracted from the other side femoral head.Results:Compared to the control and Youguiyin group,femoral head from experimental group changed extraordinary on histopathology,while the change was not so significant between other joints or position.Conclusion:At the same time of SANFH modeling successful,other joints or position were affected,Knee affected more obviously.The process of SANFH is complex.There are a lot of signals and pathways impact the process,and the number of proteins involved is enormous.Youguiyin regulats multiple biological processes of SANFH.The idea of further study about pharmacodynamic mechanism can be extended.Youguiyin efficacy study on SANFH enrich the theory of TCM "Drug toxicity loss of kidney osteoclasts" " kidney yang ".
