located the first place in the incidence and death rate of all kinds of tumors in China.The occurrence of gastric carcinoma is a multiple factors,multiple steps process that involves various mechanisms.At present,it is considered that the pathogenesis of gastric carcinoma relates to TGFβ/smads signaling.Many studies indicate that disruption of TGFβ-signaling due to alteration in any member of TGFβ/smads signaling pathway leads to carcinogenesis and development of gastric cancer.By comparing the expression of TGFβ1,Smad3,Smad7 and ki-67 in gastric cancer and normal gastric mucosa tissues and their relationship with the clinical pothology characteristics(differentiation,metastasis of lymph nodes)of gastric carcinoma,they might provide theoretical basis to the study of the pathogenesis of gastric carcinoma and new targets of clinical treatment.Part Ⅰ The relationship between TGFβ1,Smad3,Smad7,ki-67 and gastric carcinomaObjectiveTo detect the expression of TGFβ1,Smad3,Smad7 and ki-67 in normal and carcinomatous gastric tissue by immunohistochemisty and RT-PCR,and reveal the relationship between TGFβ/smad and gastric carcinomaMethodsTo collect 60 cases of gastric carcinoma and 20 cases of normal gastric tissue(away the tumor more than 10 centimetres)which were surgically removed and certified by pathology in Chengde Medical College affiliated hospital from December,2010 to September,2011.Immunohistochemisty was adopted to detect the expression of TGFβ1,Smad3,Smad7 and ki-67 in normal and carcinomatous gastric tissue;the expression of TGFβ1mRNA,Smad3mRNA,Smad7mRNA and ki-67mRNA was detected by RT-PCR in normal and carcinomatous gastric tissue.To analyse the experimental data we combined clinical pothology with statistics.Results1.1 The expression of TGFβ1 Smad7 and ki-67 in carcinomatous gastric tissue were higher than they were in normal gastric tissue(P<0.05).The expression of Smad3 in carcinomatous gastric tissue was lower than that in normal gastric tissue(P<0.05)1.2 The expression of TGFβ1,Smad7 and ki-67 in gastric carcinoma which were well differentiated and moderately differentiated were lower than they were in poorly differentiated gastric carcinoma(P<0.05).The expression of Smad3 in gastric carcinoma which were well differentiated and moderately differentiated was higher than that in poorly differentiated gastric carcinoma(P<0.05).1.3 The expression of TGFβ1,Smad7 and ki-67 in gastric carcinoma without lymph node metastasis were lower than they were in gastric carcinoma with lymph node metastasis(P<0.05).The expression of Smad3in gastric carcinoma without lymph node metastasis was higher than that in gastric carcinoma with lymph node metastasis(P<0.05).2.1 The expression of Smad3mRNA in carcinomatous gastric tissue was lower than that in normal gastric tissue(P<0.01).The expression of TGFβ1 mRNA,Smad7mRNA and ki-67mRNA in carcinomatous gastric tissue were higher than they were in normal gastric tissue(P<0.01).2.2 The expression of TGFβ1mRNA,Smad7mRNAandki-67mRNA in gastric carcinoma which were well differentiated and moderately differentiated were lower than they were in poorly differentiated gastric carcinoma(P<0.01).The expression of Smad3mRNA in gastric carcinoma which were well differentiated and moderately differentiated was higher than that in poorly differentiated gastric carcinoma(P<0.01).Conclusions1.Immunohistochemical positive signals of TGFp1,Smad3,Smad7 and ki-67 were significant differences between normal gastric tissue and gastric carcinoma,also were concerned with differentiation degree and lymph node metastasis of gastric carcinoma.2.TGFβ1Smad3mRNA,Smad7mRNA and ki-67mRNA were significant differences between normal gastric tissue and gastric carcinoma,also were concerned with differentiation degree of gastric carcinoma.Part Ⅱ The relationship amongTGFβ1,Smad3,Smad7 and ki-67 in the gastric carcinomaObjectiveTo research the relationship among TGFβ1,Smad3,Smad7 and ki-67 in carcinoma through analyzing experimental data of them.MethodsTo analyse experimental data of TGFβ1,Smad3,Smad7 and ki-67 by the statistical analysis software.Results1.The correlation coefficient of TGFpland Smad7is:r=0.286(P<0.01);the correlation coefficient of TGFβand Smad3 is:r=—0.330(P<0.01);the correlation coefficient of Smad3and smad7is:r=-0.425(P<0.01).The correlation coefficient of TGFβland ki-67 is:r=0.186(P<0.01);the correlation coefficient of Smad7and ki-67is:r=0.201(P<0.01);the correlation coefficient of Smad3and ki-67is:r=-0.547(P<0.01).2.The correlation coefficient of TGFβimRNA and Smad7mRNA is:r=0.67(P<0.01);the correlation coefficient of TGFp1mRNA and Smad3 mRNA is:r=—0.47(P<0.01);the correlation coefficient of Smad3 mRNA and smad7mRNA is:r=—0.42(P<0.01).The correlation coefficient of TGFβ1 mRNA and ki-67mRNA is:r=0.48(P<0.01);the correlation coefficient of Smad7mRNA and ki-67mRNA is:r=0.51(P<0.01);the correlation coefficient of Smad3 mRNA and ki-67 mRNA is:r=—0.39(P<0.01).ConclusionsThe correlation of TGFβ1and Smad7was positive;The correlation of TGFβand Smad3was negative;The correlation of Smad3and Smad7was negative;The correlation of ki-67and TGFβ1was positive;The correlation of ki-67and Smad7was positive;The correlation of Smad3and ki-67was negative in gastric
