Objective To clarify the cross link between EMT process and TGF?1,smad3 and smad7 in both tumor tissue and serum.To verify the EMT process,we investigated e-cadherin and Vimentin protein expression in the resected tissues of colorectal cancer or colon polyps.And smad3,smad7 were detected to evaluate the activation of TGF?1/smads pathway.ELISA assay were used to detect the content of TGF?1,smad3 and smad7 on patients with colorectal cancer,intestinal polyp patients and healthy volunteers.While combined with these clinical datas,the significance of serum free TGF?1,smad3 and smad7 were assessed.Methods This study enrolled 136 patients who was firstly diagnosed with colorectal cancer and never receive any anti cancer therapy before from February 2012 to September 2012 in gastrointestinal surgery department and gastroenterology department in the first renmin hospital of huai nan city.94 Patients with colon polyps who hospitalize at same time were enrolled as control group and 60 healthy adults were used as blank group.The resected or endoscopic biopsy specimens from experimental group and the control group were fixed and embedded by paraffin.Immunohistochemistry stain(IHC)were used to detect the expression of E-cadherin and Vimentin.The blood samples should be collected from both the experimental group and the control group before they received any treatment,and samples from the health adult volunteers were also collected.Enzyme-linked immunosorbent assay(enzyme linked immunosorbent assay,ELISA)were used to detect content of TGF?1,Smad3 and Smad7.The data in this study were statistic analyzed by the SPSS22.0.The IHC staining was graded and ranked according to the staining strength and the positive cell proportion.The ELISA results were statistical stand by the mean ± standard deviation(x ± s).Ranked data were analyzed by rank sum test and Spear-man correlation test.Measured data were compared with analysis of variance.All of the above analyses were given P < 0.05 as significance.Results The expression of E-cadherin in the experimental group was lower than that in the control group(P < 0.01),and the expression of Vimentin was higher than that in the control group(P < 0.01).ELISA results showed that the expression levels of TGF?1 and smad3 in the experimental group were significantly higher than that in the control group and the blank group(P < 0.01),and the expression level of smad7 was significantly lower than that in the control group and the blank group(P < 0.01).There was no statistical difference of TGF?1 and Smad3 in serum between the blank group and the control group.Tthe content of TGF?1and Smad 3 in patients with Dukes C and D stage were higher than those in Dukes A and B group,while the content of smad7 decreased.Conclusion EMT process happpen in the process of colorectal cancer,and that were associated with TGF?1/smads pathway activation.The serum content of TGF?1,smad3 and smad7 were elevated and related to Dukes stage.The increase of TGF?1,smad3 and smad7 in serum can reflect,in a certain extent,the activation of TGF?1/smads pathway in tumor tissue.And this corss-link may play an important role in the process of the development of colorectal cancer.
