Purpose:This research aims to construct an eukaryotic expression vector containing IER3 gene.Methods:The IER3 gene was amplified from human non-small cell lung cancer cells A549 by PCR.After confirmed by sequencing the pGEMT easy vector,IER3 gene was subcloned into pcDNA3.1-6His vector.Then the pcDNA3.1-6His-IER3 was transfected into A549 cells and observed the expression of target protein by Western blot and fluorescence microscope.Results:The IER3 gene was inserted into pcDNA3.1-6His vector correctly,and was expressed in the A549 cells confirming by Western blot.In addition,the IER3 gene localized in the cytoplasm.Conclusion:IER3 eukaryotic expression vector was successfully constructed.The research provides a basis for exploration of IER3 function in the tumororigenesis and cancer progression... |
