Objectives:To observe the lipid-lowering effect and the antioxidation of inulin oligosaccharides and to explain their effect mechanism in hyperlipidemia C57BL/6J mice induced by high fat diet.Methods:1.C57BL/6J mice aged 8 weeks,were randomly divided into normal group,model group,inulin oligosaccharides high,medium and low dose group,positive drug lovastatin group,with 10 mice in each group.After adaptive feeding for a week,normal group was given normal diet,the other groups were fed on atherosclerosis exclusive high fat diet.Drug groups were given drugs by intragastric administration once a day,normal group and the model group were lavaged with physiological saline once a day.The mice were fed for 10 weeks.2.Mice were ablated the eyeball,and the serum was separated.Test the level of TC,TG,LDL-C and HDL-C.3.Detected the level of MDA,CAT,GOT and T-AOC in the liver.4.Scanning electron microscope was used to observe the mice artery endothelial changes.5.HE sections were made to observe the effects of inulin oligosaccharides on fatty liver pathological changes in mice.6.Transmission electron microscope was used to observe fatty liver pathological changes in mice.7.Elisa method was used to test the level of Lp(a)、apoA1 and apoB.7.Western blot method was used to detect the NF-κB,CRP and PPARγ expression in the liver.8.Immunohistochemical method was used to detect the NF-κB expression in the liver.Results:Comparing to the normal control group,mice in the model control group had significant higher level of TG,TC,LDL-C,apoB,Lp(a),and significant lower level of HDL-C,apoAl in the serum;comparing to the model control group,mice in the inulin oligosaccharides group had signifant lower level of TG,TC,LDL-C,apoB,Lp(a)and significant higher level of HDL-C,apoAl.Comparing to the normal control group,mice in the model control group had significant higher level of MDA,GOT,and significant lower level of T-AOC,CAT in the serum;comparing to the model control group,mice in the inulin oligosaccharides group had signifant lower level of MDA,GOT and significant higher level of T-AOC,CAT.Comparing to the normal control group,mice in the model control group had significant higher level of NF-κB,CRP,and significant lower level of PPAR-y in the serum;comparing to the model control group,mice in the inulin oligosaccharides group had signifant lower level of NF-κB,CRP,and significant higher level of PPARy.Conclusion:Inulin oligosaccharides can protect the vascular endothelium and lessen the lesions in the liver of hyperlipidemia mice.Also,decreasing serum TG,TC,LDL-C,apoB,Lp(a)levels and increasing HDL-C,apoAl level in serum can be detected in oligosaccharides group,Otherwise,increasing T-AOC,CAT levels and the decreasing MDA,GOT levels can be observed.Comparing to the normal control group,mice in the inulin oligosaccharides group had significant lower NF-κB and CRP expression and higher PPARy expression.These results indicated that the mechanism of inulin oligosaccharides on lipid-lowering and AS resistant was resistance to oxidative stress and activation of PPARy,which affect the inflammatory mediators of CRP and NF-κB pathway.
