| AIM:To investigate the inhibitional effect of Stichopus japonicus acidic mucopolysaccharide(SJAMP)on cell proliferation in pancreatic carcinoma cell line SW1990,and to discuss the possible mechanism through Hippo pathway.METHODS:We use semi-quantitative RT-PCR to check the mRNA expression of YAP、MST1、Survivin、Caspase-9、TEAD1 in SW1990 cells after SW1990 cells were cultivated in 8mg/ml SJAMP for 24,48,72 h.The protein expression of YAP and phosphorylated YAP(p-YAP)in SW1990 cells was detected by Western blotting.RESULTS:8 mg/ml SJAMP inhibited mRNA expression levels of Hippo pathway relevant gene YAP,TEAD1 and apoptin survivin(F=8.36,P<0.01;F=5.87,P<0.05;F=4.81,P<0.05)and up-regulated Caspase-9、MST1 mRNA expression(F=10.66,P<0.01;F=6.56,P<0.05)both in a time-dependent manner.Western blot analysis showed that SJAMP reduced the level of YAP protein expression(F=22.96,P<0.01)and enhanced the the level of pYAP protein expression in a time-dependent manner(F=55.88,P<0.01).CONCLUSION:SJAMP inhibits the proliferation of pancreatic carcinoma cells perhaps by activating Hippo pathway and regulating the relevant genes. |
