Objective To observe the effect of Shenfu Injection on the Cardiomyocyte Apoptosis and the Expression of Bcl-2,Bax in Rat with Sepsis,which to provide evidence for clinic.Methods The sepsis model was established by cecal ligation and acupuncture.A total 24 male Sprague-Dawley rats were randomly divided into sham operation group(Sham group),sepsis group(CLP group)and Shenfu injection treatment group(SFI group)(each n=8).In CLP group the rats were injected with 5mL/kg saline into their peritoneal cavities at 0 hour,6 hour and 12 hour after CLP.The rats in Sham group were treated the same way except no ligation and puncture of the cecum.In SFI group the rats were treated with 5 mL/kg Shenfu Injection fluid into their peritoneal cavities at 0 hour,6 hour and 12 hour after CLP.The level of Creatine phosphokinase(CK),Creatine kinase isoenzyme(CK-MB)and cardiac troponin I(cTnI)was observed pathological changes in cardiac muscle was observed by HE dyeing,Myocardial apoptosis was detected by optical microscopy technology and TdT-mediated dUTP Nick End Labeling(TUNEL).mRNA expression levels of Bcl-2 and Bax gene were measured by reverse transcriptional polymerase chain reaction(RT-PCR).Results 1.The myocardial apoptosis index in CLP group and SFI group were higher than that in Sham group(P<0.05),while that in SFI group was lower than that in CLP group(P<0.05).2.The level of CK-MB,CK and cTnI in CLP group and SFI group were higher than that in Sham group(P<0.05),while that in SFI group was lower than that in CLP group(P<0.05).3.The results of RT-PCR:Compared with Sham group,the expression of Bcl-2 mRNA levels in CLP group and SFI group were significant decreased(P<0.05)and Bax mRNA were significant increased(P<0.05).Compared with CLP group,the expression of Bcl-2 mRNA levels in SFI group were significant increased(P<0.05)and Bax mRNA were significant decreased(P<0.05).Conclusion Shenfu injection could reduce apoptosis in the septic rats myocardium by increase the Bcl-2 mRNA expression and decrease the Bax mRNA expression.
