Objective To investigate the serum levels of signal transducer and activator of transcription-4(STAT-4) and signal transducer and activator of transcription-6(STAT-6) in healthy controls and recurrent spontaneous abortion (RSA) patients. Discuss the relationship between the incidence of RSA and STAT4/STAT6 to provide evidence-based diagnose and treatment for RSA.Methods The concentrations of serum levels of STATs in healthy controls and recurrent spontaneous abortion (RSA) patients were measured by ELISA kit. Median serum STATs concentrations among different groups were compared by Mann-Whitney U Non-parametric test or t test for their significance of difference. The correlation between STAT4 and STAT6 protein expression is analyzed by Spearman correlation method.Results1.The level of STAT4 in normal early pregnancy is 199±48pg/ml. This level in RSA patients is 281±60pg/ml. The result in RSA patients is significantly higher (P<0.001, Z=-4.648)2.The level of STAT6 in normal early pregnancy is 288±67pg/ml. This level in RSA patients is 143±57pg/ml. The result in RSA patients is significantly lower (P<0.001, t=9.040) 3.The ratio of STAT4/STAT6 in normal early pregnancy is 0.7680±0.3847. This ratio in RSA patients is 2.7719±3.0382. The result in RSA patients is significantly higher (P<0.001, Z=-5.681)4.The level of STAT4 in RSA patients before their treatments is 281±60pg/ml, this level after their treatments is 165±60pg/ml. The result in RSA patients after their treatments is significantly lower (P<0.001, Z=-7.259)5.The level of STAT6 in RSA patients before their treatments is 143±57pg/ml, this level after their treatments is 308±48pg/ml. The result in RSA patients after their treatments is significantly higher (P<0.001, t=-15.508)6.The ratio of STAT4/STAT6 in RSA patients before their treatments is 2.7719±3.0382, this ratio after their treatments is 0.5699±0.2665. The result in RSA patients after their treatments is significantly lower (P<0.001, Z=-8.417)7.Measured by Spearman correlation analysis, the serum level of STAT4 and STAT6 are significant correlated (P<0.001), Correlation Coefficient=-0.625,-0.698, negative correlation.Conclusions1. STAT4 protein expression dominated in recurrent spontaneous abortion patients while STAT6 was inhibited. This may be one of the pathogenic mechanisms in recurrent spontaneous abortion. 2. STAT6 protein expression increased while STAT4 decreased after treatments were given to recurrent spontaneous abortion patients. This could be the indicators of clinical treatment outcomes.3. Negtive correlation between STAT4 and STAT6 proteins existed. The increased expression of STAT4 protein may inhibites the STAT6 signaling pathway.
