Studies On The Effective Components And The Establish Of Quality Standard Of Cistanche Salsa

Objective:To Study reference substance of cistanoside A from Cistanche salsa,and determine the purity of it at the same time;To establish quality standard of Cistanche salsa;To investigate in vitro antioxidative acyivities of three kinds of high phenethyl alcohol glycoside content?echinacoside,cistanoside A,acteoside?.Methods:1.Establishing the preparation technology of cistanoside A;Authenticating the chemical structure of cistanoside A by UV,IR,HPLC-MS and NMR;Testing the purity of cistanoside A by physical constant,TLC and HPLC;Measurig the purity content of cistanoside A by area normalization method,HPLC.2.Testing the characters,identifications,inspection,and the ethanol soluble extractive,based on the Chinese Pharmacopeia 2010;Establishing the HPLC on the assaying of echinacoside,cistanoside A,acteoside as reference substance.3.Investigated the reducing power and radical?·OH,·O₂^-and DPPH·?scavenging effect of echinacoside,cistanoside A,acteoside by spectrometer?UV?.Results:1.Having established the preparation technology of cistanoside A which was from n-BuOH extract,which was separated from Cistanche salsa,and it was about 95.5%pure which has been measured by area normalization method,HPLC.2.Cistanche salsa?11 batches?which have been tested were that,they contains6.48%¹0.06%of moisture,7.03%¹1.53%of ash,0.30%¹.69%of acid insoluble ash,45.93%⁶2.61%of ethanol soluble extractive,heavy metals which was not more than0.005‰;arsenic which was not more than 0.001‰;echinacoside which was0.22%⁶.03%,cistanoside A which was 0.09%¹.55%,and acteoside which was0.08%¹.28%by HPLC.3.The results of the reducing power and radical?·OH,·O₂^-and DPPH·?scavenging effect of echinacoside,cistanoside A,acteoside,which have been investigated by spectrometer?UV?,were that acteoside>echinacoside>cistanoside A.Conclusion:1.Cistanoside A,the reference substance which were separated during the experimentation,could provide reference material for farther controlling the quality standard of Cistanche salsa.2.Quality standard of Cistanche salsa,which has been established,provided important basis for the quality evaluation of Cistanche salsa,according to many-sided index of quality evaluation and quality standard of Cistanche deserticola which has been established,in the Chinese Pharmacopeia 2010.3.Antioxidant activity on three kinds of phenethyl alcohol glycoside,which has higher content of Cistanche salsa,which has been researched,laid foundation for further study on antioxidation of phenethyl alcohol glycoside from Cistanche salsa.
