At present,more attention was paid to multi-drug resistant Pseudomonas aeruginosa infection which has lead to the higher mortality rate and the less application of targeted drug.After years of clinical trials,doctors confirmed that most of the traditional Chinese medicines would inhibit the infections of drug-resistant,and can be reversed the bacterial resistance.Qi-gui-yin(QGY)is a traditional Chinese patent medicine,who is composed of five herbal medicines,Astragalus membranaceus(Fisch.)Bge.,Angelica sinensis(Oliv.)Diels,Lonicera japonica Thunb.,and Artemisia annua L.,Polygonum cuspidatum Sieb.et Zucc..It can be used for the treatment of multi-drug resistant Pseudomonas aeruginosa infection.Currently,more than 3000 cases have been used in clinical and the results are satisfactory.However,we have no accurate data to elaborate the bioactive ingredients in QGY comprehensively,which is essential to explain the mechanism.First,we classified and summarized the chemical composition in five single herbs,and then established the LC-MS method to analyze the chemical composition in QGY and drug serum,urine and feces.Therefore,a system serum pharmacochemistry study based on LC-MS to elaborate the bioactive ingredients in QGY was performed as follows:1?Summarized of chemical composition in QGYBy reading literatures,we classified and summarized the chemical composition and their drug activity in five single herbs of QGY.Flavonoids,saponins and polysaccharides are mainly chemical composition in Astragalus.Angelica containing volatile oil,organic acids and phenols.The chemical composition of Honeysuckle are organic acids,flavonoids and volatile oils.The chemical composition of Artemisia annua are flavonoids,coumarin,volatile oil and sesquiterpenes.Polygonum containing quinones,flavonoids and phenolic.Among them,the pharmaceutically active of flavonoids are antibacterial and anti-inflammatory.The pharmaceutically active of saponins,quinones,organic acids,volatile oil and polysaccharides are anti-inflammatory,antibacterial,antibacterial,antibacterial and anti-virus.2?The study of chemical composition in QGY by LC-ESI-MS/MSA LC-ESI-MS/MS method for identifying chemical composition in QGY was developed.15 compounds were identified in QGY with the developed method by analyzing the retention time,molecular weight and cleavage pathway,including flavonoids,phenylpropanoid,anthraquinones and other compounds.The result showed that the developed method is applicable to identifying the complex chemical composition in QGY.3?The study of chemical composition in QGY by LC-ESI-HR-MS/MSOn the basis of the developed method,we identified the chemical composition in QGY with LTQ-Orbitrap.By comparing the retention time,accurate molecular weight and MS/MS fragmentation with standards and related literatures,60 compounds were identified or tentatively characterized in QGY with HPLC-ESI-LTQ-Orbitrap,including flavonoids,phenylpropanoid,anthraquinones,isoflavans,isoflavones,saponins and other compounds.The result elaborated the chemical composition in QGY comprehensively.4?The study of chemical composition in serum and metabolites containing QGYHPLC-ESI-LTQ-Orbitrap was used to investigate the chemical composition in serum,urine and feces.A total of 13 compounds were identified in the serum,18 compounds in urine,3 compounds in feces,including flavonoids,phenylpropanoid and anthraquinones.Among them,11 compounds,metabolites were found in the urine except Luteolin and 5,7,4'-trihydroxy flavonol-7-O-?-D-glucopyranoside.In our study,the chemical composition in serum containing QGY were identified and corroborated by the urine and feces.Finally,the bioactive ingredients in QGY were elaborated preliminarily.
