Objective To study the effects of Bushen Zhuanggu Granules on the quality of life and micro-structure and biomechanical properties of the femur of SAMP6 mice,and the mechanism of its pharmacodynamics was discussed by detecting serum hormone and bone metabolism index,and to infer that the pharmacodynamics and mechanism of Bushen Zhuanggu granule on senile male osteoporosis and explore whether it would replace growth hormone for the treatment of osteoporosis in elderly men from the study.Methods 30 SAMP6 aged three mouths were divided randomly into model group,growth hormone?GH?group and traditional Chinese medicine?TCM?group,10 normal accelerated male mice?SAMR1?with the same age were selected as homologous control group.After a week of normal feeding,the mice of GH group and TCM group were given medicine,weighed once a week and we adjusted dose by the weight.The mice of model and normal groups were given equivalent normal saline,and the living conditions of mice in each group were observed and recorded every day,and the living conditions of mice in each group were observed and recorded every day.After 3 months,5 mice in each group were randomly selected to take blood in a 1.5 ml centrifuge tube from their eyeballs under anesthesia,centrifuged?25 degrees,3000rpm,15min?when the blood was completely solidified at normal temperature,and kept in the refrigerator at-80 C,to be used to detect the content of serum GH,IGF-1,T,E2,BGP,CTX-1 and PINP.To take out the bilateral femur after being executed with the method of cervical spondylolisthesis,the left femur for measuring the bone mass and femoral geometry size and mechanical geometry of bone section and biomechanics capabilities detection,the right femur for Micro-CT detection.Result Compared with the normal group,the quality of life was decreased,such as darker hair,irascibility etc,and significant weight gain;the cortical bone of femur was thinned,biomechanical properties and bone density was significant decline,bone microstructure presents osteoporosis in SAMP6 mice,and the results of SAMP6 mice were more obvious in 6 months than 3 months.The levels of GH,IGF-1,E2,T and PINP in serum were significantly lower than those in normal controls?p<0.05or p<0.01?and CTX-1,BGP were significantly increased.?P<0.05 or P<0.01?,showed that the bone formation of SAMP6 mice decreased and the bone resorption increased.Compared with the model group,the quality of life of SAMP6 mice in TCM group was improved without any other side effects.3 months later,cortical bone thickness and sectional area increased?p<0.05?,the diameter of the bone marrow cavity and the cross sectional area decreased?p<0.05?,the parameters of bone structure mechanics were significantly improved?p<0.01?,the maximum stress and elastic stress increased?p<0.01?,the stiffness coefficient increased?p<0.05?,the BV/TV,Tb.Th,Tb.N and Conn.Dn increased,Tb.Pf,Tb.Sp,SMI and DA decreased,and the difference was statistically significant?P<0.05 or P<0.01?;the bone trabecula were wider,compactstructure,go in line and uniform thickness,continuity well and the mesh structure was perfect nearly,the level of serum GH,IGF-1,T increased and BGP decreased?p<0.05 or p<0.01?,PINP and E2 increased?P>0.05?of the SAMP6 in TCM group;6months later,the cortical bone thickness and cross-sectional area increased?p<0.01?,bone marrow cavity diameter and cross-sectional area decreased?p<0.05?,the mechanical geometry of bone section were significantly improved?p<0.01?,the maximum load and elastic load increase?p<0.05?,the maximum,maximum bending moment,deflection,energy absorption,elastic stress and stiffness coefficient increased?p>0.05?,the elastic modulus decreased?p>0.05?,bone mineral density increased?p>0.05?,the BV/TV,Tb.Th,Tb.N and Conn.Dn increased,Tb.Pf,Tb.Sp,SMI and DA decreased?P<0.05 or P<0.01?,the bone microstructure was also improved,but the bone trabecula were thinner and poor alignment,the reticular formation began to break and there is no bone trabecular in the bone marrow zone,the cortical bone layer became thinner,showing mild osteoporosis,the level of serum GH,T,PINP increased and CTX-1,BGP decreased?p<0.05 or p<0.01?,IGF-1and E2increased?P>0.05?of the SAMP6 in TCM group.Compared with growth hormone group,there had no the conditions reducing the quality of life,such as grumpy,aggressive,genital erosion etc,in TCM group;3 months later,SMI decreased and the level of T was significantly increased?P<0.05?,there was no significant difference in other parameters?p>0.05?;6 months later,cortical bone thickness and cross sectional area increased significantly?p<0.01?,the geometry of bone cross-section is increased?p<0.05?,BV/TV,Tb.N and Conn.Dn increased and Tb.Sp,SMI and DA decreased?P<0.05 or P<0.01?,there was no significant difference in other parameters?p>0.05?.Conclusion Bushen Zhuanggu granules could improve the quality of life of SAMP6 mice,promote bone formation and inhibit bone resorption by regulating the secretion of IGF-1,T and E2 and the levels of bone metabolism index?BGP,CTX-1,PINP?,and then improve the mechanical response function to macroscopic geometric structure and the micro-structure and bio-mechanical properties of bone of SAMP6 mice,increase its bone toughness and strength,and improve its resistance to external forces deformation and fracture,and then prevent and treat the osteoporosis and reduce the risk of fracture;it could increase bone mass in SAMP6 mice with long term administration of Bushen Zhuanggu granules;It follows that Bushen Zhuanggu granules could adjust the GH-IGF-1 axis and the secretion of T,improve bone metabolism,promote bone formation and inhibit bone resorption,and then improve the bio-mechanical properties of bone and the number and quality of trabeculae,and then prevent and treat the osteoporosis in elderly men and reduce the risk of fracture;Compared with the results of the effect of growth hormone,we could see that there is hope of Bushen Zhuanggu Granules to replace growth hormone,which is expensive and has more side effects,to treat osteoporosis in elderly men.
