Aim:This project belongs to the field of traditional Chinese medicine,the development and application of new drug preparations of Chinese medicine research.This topic on the basis of the theory of Chinese medicine kidney advocate bone combined with modern science and technology,give full play to advantages of Chinese medicine,by observation of basic and clinical indicators,for older women osteoporosis more comprehensive system of research,reveals the treatment of TCM and the mechanism of the older women osteoporosis,explore kidney raw pulp,supplemented by yiqi huoxue for prevention of elderly women osteoporosis and unique new method effectively.And according to the prescription composition of the preparation of the preparation process,and to develop the quality standards for the preparation.Observed in kidney marrow method on the basis of preparation of Duzhong Zhuanggu granula strong bone granule on osteoporosis(OP)research on the effects of bone mineral density of bone metabolism,female mice from biological molecular level to discuss the OP pathogenesis and benefiting kidney marrow function mechanism of traditional Chinese medicine to prevent and control the OPMethod:According to the composition of the prescription,three factors and three levels L9(34)orthogonal experiment was used to investigate the water addition,decoction time and decoction times in the water decoction process,and the optimal water extraction process was optimized by comprehensive score of extraction rate and puerarin extraction amount under different conditions.According to the optimal water extraction process of water decoction and alcohol precipitation process,and different alcohol precipitation conditions of the extraction rate and puerarin extraction amount as evaluation index to optimize the alcohol precipitation process,the alcohol precipitation of water extract concentrated to the relative density of 1.25-1.30(60℃)extract,choose different proportion of auxiliary materials mixed granulation,investigate the optimal molding process to make particles.Clean the ovariectomized adult female mice 60,were randomly divided into the intervention of traditional Chinese medicine group,western medicine control group,blank control group,model group,each group of 15 only,and were given the duzhong zhuanggu granula,Calcium Carbornate and Vitamin D3 Granules,saline gavage administration.After 16 weeks of serum,serum Ca,automatic biochemical analyzer P,BGP,BALP concentration in the femur bone mineral density was measured by dual energy X-ray absorptiometry,and observe the changes in the structure of bone tissue under the microscope.Experimental results analyzed with SPSS 16.0 software,the indicators results to mean +/-standard deviation(+/-S)said,T test,P values,P<0.05 was statistically significant.Result:The traditional Chinese medicine group and Western medicine control group after 16 weeks of treatment,compared with the model group significantly increased serum Ca,P and the density of bone(femur,spine and whole body)expression level(P<0.05);the traditional Chinese medicine intervention group(BALP and bone density of femur,spine and whole body)on the transfer rate is greater than the western medicine group,while BGP was significantly reduced(P<0.05).The western medicine control group,traditional Chinese medicine intervention group after 16 weeks of bone tissue morphology measurement and model group(p<0.05)were significantly different,significantly higher than the model group,there was statistical significance.The western medicine group,Chinese medicine group(p>0.05),no significant difference was not statistically significant.Conclusion:Bones of the bark of Duzhong Zhuanggu granula on the OP female increase bone density concentration of serum Ca,P,inhibition of BGP,change the bone structure,so as to promote bone formation,to slow to female ovary development and incidence of osteoporosis have certain effect.
