Objective:Incontinence-Associated Dermatitis,whether in the community,nursing homes,long term care facilities,or in the clinical departments of the hospital,intensive care,all have a very high incidence of Incontinence-Associated Dermatitis occurs to the patient brought great pain and increased burden on the economy.Therefore,the prevention and treatment of Incontinence-Associated Dermatitis is a hot issue that we should study.We studied the establishment of a rat model of Incontinence-Associated Dermatitis through different methods.Based on animal experiments,we used Topical oxygen therapy with different flow rates to intervene in incontinence rat models to observe the effect of topical oxygen therapy on Incontinence-Associated Dermatitis.The aim is to find an easy-to-replicate method for establishing a rat model of incontinent dermatitis and to study the effect of Topical oxygen therapy on the healing of Incontinence-Associated Dermatitis,and to provide experimental evidence for further application of Topical oxygen therapy to the clinical control of incontinent dermatitis.Methods: 1.The establishment of rat models of Incontinence-Associated DermatitisEighteen 8-week-old SD male rats were divided into three groups of 6 mice each.The three groups were normal saline group,artificial urine group and trypsin solution group.The method of establishing consistent with previous steps Incontinence-Associated Dermatitis rat model,are all impregnated on the back of rats with a cotton ball,that the distinction between different groups selected cotton ball soaked in the solution.First,the rat's back was skinned and the hair removal treatment was completed.The second step was to cover the cotton balls on the back of the rats: The corresponding solutions of the cotton balls were: normal saline group,artificial urine group,and 4g/100 ml trypsin solution group.The third step was to fix the cotton ball on the back of the rat: it was fixed with a medical PU film waterproof tape and fixed again with LAC elastic muscles.The fourth step is to fill the cotton ball: After fixing the cotton ball,the corresponding solution is injected into the rat's back cotton ball according to grouping each time on the afternoon and afternoon to keep the cotton ball moist.In the fifth step,the dermatitis was observed.After three days,the tape was disassembled,and the dermatitis was observed macroscopically.The area of dermatitis was measured daily and the severity of dermatitis was scored until the dermatitis condition healed and the time for dermatitis healing was recorded.2.Topical oxygen therapy intervention of incontinent dermatitis rat modelThirty-two 7-8 week-old SD male rats were divided into 4 groups with 8 in each group.The four groups were oxygen therapy low flow group,oxygen therapy middle flow group,oxygen therapy high flow group,and blank control group.First,the back of rats modeling on rats after modeling assessment of the severity,extent to ensure that each rat were severe dermatitis.The second step is to perform partial oxygen therapy: Place the rat in a compact bag,expose the rat's fore-paw and head,and insert the oxygen tube from the side of the sealing mouth.Place the oxygen tube on the back of the rat for the right dermatitis site.The compact bag double seals are squeezed tightly and compacted to seal the tape,and the seal is pasted with tape to increase the tightness.The third step is to perform Topical oxygen therapy: one hour of Topical oxygen therapy is performed every day,Topical oxygen therapy is applied for 3 days,and dermatitis samples are taken after 3 days for severity assessment,dermatitis healing rate calculation,and epithelial histopathological observation and evaluation.The Topical oxygen therapy oxygen flow rate was set to 3L/min in the low-flow oxygen therapy group,6 L/min in the oxygen therapy mid-flow group,and 9L/min in the oxygen therapy high-flow group.Blank control group: After the modeling is successful,no treatment is performed.Results: 1.The establishment of rat models of Incontinence-Associated DermatitisThe trypsin group scored far more than the other two groups in terms of the scale of dermatitis modeled or dermatitis severity.The same dermatitis recovery time is also much greater than the other two groups.The healing time of dermatitis recovered between 6-12 days with an average of 8.83 days(P<0.001).Although artificial urine group can also establish dermatitis model,the area of dermatitis established is relatively narrow.The degree of dermatitis established is relatively light.Some rats showed reddish and reddened skin.There is no rash or breakage of the skin and it can be restored in 2-5 days.In the model established by the saline group,only part of the rat's skin rose and whitening appeared.The severity of dermatitis is very light and can be restored one day after the stimulus is removed.2.Topical oxygen therapy intervention of incontinent dermatitis rat modelFour groups of incontinent dermatitis rat models were treated with Topical oxygen therapy.The total area of dermatitis was smaller than before.Most of the rat dermatitis develop scarring as it develops.The dermatitis score was also lower than before.All have different degrees of dermatitis healing.Two pairs of data from the two groups were compared.In terms of healing rate,dermatitis severity,dermatitis section score,and inflammatory cell infiltration,the oxygen therapy high flow group,oxygen therapy middle flow group,and oxygen therapy low flow group,blank control group,the difference between the two groups Statistically significant(P<0.05).However,there was no significant difference between the two groups(P>0.05).There was no significant difference in the data between oxygen therapy low flow group and blank control group(P>0.05).Conclusion:1.The concentration of 4% trypsin solution and artificial urine and saline group as a comparison can be successfully constructed in severe rat model of Incontinence-Associated Dermatitis.The establishment of a rat model of incontinent dermatitis is simple,convenient,and easy to replicate,and can be further promoted and further studied.2.The experimental results demonstrate from various aspects that local oxygen therapy can effectively relieve the symptoms of incontinence dermatitis.Promote the healing of incontinence dermatitis.Among them,partial oxygen therapy is best performed with an oxygen flux of 6-9 L/min.Comprehensive literature and experimental results show that local oxygen therapy is worthy of recognition not only in chronic wounds but also in stimulating wounds and incontinence dermatitis.The research results are also worthy of promotion and further research in the clinic.
