Background:Knowledge of the genetic architecture of Ischemic stroke (IS) has been quite limited. Mostsignificant associations of candidate genes with IS have been difficult to replicate.Growtharrest-specific gene 6 (Gas6) is Axl receptor tyrosine kinases family ligands, which participationin inflammation, platelet aggregation and atherosclerosis. Recent years, Single NucleotidePolymorphisms (Single Nucleotide Polymorphisms, SNPS) of genetic markers appear, whichgive an important basis of IS molecular biology in the pathogenesis of research provides.Objective:(1) To investigate the association between the rs8191974G>A and rs7400722C>Tpolymorphisms in Gas6 gene and IS.(2) To investigate the association between the haplotype of rs8191974G>A and rs7400722C>Tpolymorphisms and IS.(3) To explore and assess the possible interaction effects between the gene polymorphisms andenvironmental factors.Methods:A total of 119 patients with with IS and 217 healthy individuals were were recruited in thisstudy. Using polymerase chain reaction-restrictive fragment length polymorphism (PCR-RFLP)determin gene polymorphisms. Univariate test and multiple logistic regression models were usedto explore the association between the SNPs and IS and subtype of IS. Additionally, theinteraction between SNPs and environmental risk factors were assessed by MDR1.1.0.Results:(1) The rs8191974G>A and rs7400722C>T genotype distributions was not consistent with theHardy-Weinberg equilibrium, which was removed from further analysis.(2) There were no significant differences in the genotype (P=0.80 or 0.79), allele (P=0.57 or0.50), additive model (P=0.93 or 0.58), recessive model (P=0.51 or 0.58). (3) The haplotype (G-C 0.74, G-T 0.83, A-C 0.16, A-T 0.35) frequencies of Gas6 genepolymorphisms between the group of patients with ischemic stroke and the control group.(4) After adjustment for age, gender, and frequencies of hypertension, diabetes mellitus, andsmoking, there was no significant association of genotype, allele and haplotype at any of thepolymorphism in relation to any subtype of ischemic stroke (P>0.05).(5) 2 SNP sites genotypes, show the recessive model and single body shape and LAA andSAA subtypes no specific correlation between (P>0.05);(6) The interactions were found between rs8191974G>A polymorphism, smoking andhypertension disease.Conclusion:(1) No definite conclusion could be reached regarding the involvement of the rs8191974G>Aand rs7400722C>T polymorphism of Gas6.(2) No definite conclusion could be reached regarding the involvement of the haplotype ofGas6. However, further studies are needed to explore the complex interaction betweenenvironmental factors and Gas6 gene polymorphisms in the risk of ischemic stroke.(3) rs8191974G>A polymorphism, smoking and hypertension disease have a possibleinteraction on the presence of IS, which may result in a possible effect on stroke risk .
