Angelica sinensis and Astragalus membranaceus are two kinds of the most common Chinese traditional herbs commonly used as clinical medicine,which contains many active ingredients and have been shown to possess various pharmacological effects.Modern pharmacological studies have shown that angelica sinensis has the effect of enhancing immunity,stimulating hematopoietic,protecting liver function,scavenging oxygen free radicals,anti-tumor,anti-inflammatory and anti-aging.Astragalus membranaceus has wide pharmacological efficacies such as decompression,anti-virus,anti-radiation,anti-stress and anti-oxidative activities,etc.We applied ultra-filtering technique to intercept the extracts of higher molecular weight(more than molecular weight of 20,000)and lower molecular weight(less than molecular weight of 20,000)from angelica sinensis and astragalus membranaceus(1:5(w:w)),and individually determined the amount of effective elements of polysaccharides,flavonoids and astragaloside ? in the two different molecular weight of Guiqi ultra-filtration extracts.The higher molecular weight Guiqi ultra-filtration extract contains the higher level of polysaccharides.The other two elements flavonoids and astragaloside ? are determined the higher level in the lower molecular weight Guiqi ultra-filtration extract.To study the antioxidative activity of Guiqi ultra-filtration extracts,hydroxyl radical scavenging ability,superoxide anion radical scavenging ability and reducing power were determined and compared,respectively.The results indicated that Guiqi ultra-filtration extracts had high and multiply antioxidative activity.It was found that the antioxidative activity of the lower molecular weight Guiqi ultra-filtration extract was superior to the higher molecular weight Guiqi ultra-filtration extract.D-galactose-induced aging rats model was established to estimate the influence of Guiqi ultra-filtration extracts on the contents of Hyp in skin.The results showed that Guiqi ultra-filtration extracts could significantly increase the content of Hyp(p<0.01).This results indicated that Guiqi ultra-filtration extracts could increase Hyp contents and slow down the rate of skin aging.The influence of Guiqi ultra-filtration extracts on the liver mitochondria oxidative damage and energy metabolism were estimated.As result,Guiqi ultra-filtration extracts reduced the vitality of MAO and MDA(p<0.01),increased the activity of SOD,MDH and SDH in liver mitochondria(p<0.05,p<0.01).These results indicated that Guiqi ultra-filtration extracts might play an important function,whichreduced free radical damage to the liver mitochondria and protected the normal metabolism of mitochondria.The effcet of Guiqi ultra-filtration extracts on the expression of P53 in kidney were determined.The experiment results indicated that Guiqi ultra-filtration extracts down-regulated the expression of P53 in kidney(p<0.05).The results proofed that the signal transduction pathways of P53 may play an important role in the delay of cellular senescence process.The influence of Guiqi ultra-filtration extracts on the activity of telomerase in sexual organ were analyzed.The results showed that Guiqi ultra-filtration extracts could activate vitality of telomerase(p<0.01).
