Angelica sinensis (Oliv.) Diels and Astragalus membranaceus (Fisch) Bge. are the most common lacal medical plants in Gansu Province, China. Modern pharmacological studies have shown that Angelica has the effect of anti-tumor, protecting liver function, scavenging oxygen free radicals and anti-inflammatory by activating complement system, enhancing immunity and stimulating hematopoietic function. Astragalus has a variety of pharmacological efficacies, such as cardiac and decompression, regulating blood sugar level, anti-stress, anti-virus, anti-radiationand anti-oxidation. Polysaccharides are biological macromolecules with a wide range of biological activity. It exists in nearly100species of herbs. As a new medical ingredients with higher efficiency and lower toxicity, polysaccharide attracts more attention and has become a hot field of view. In this study, Guiqi polysaccharide was prepared by optimizing ratio of Angelica and Astragalus. Furthermore, biological activities in vitro (including anti-oxidation, antibacterial, antitumor and antiviral effects) and pharmacodynamics in vivo (anti-aging, inhibition tumorsgrowth and protect mice from acute liver impairment) were studied to clarify thepharmacological mechanisms of Guiqi polysaccharide.Extraction ratio of Guiqi polysaccharide was served as target to choose factors andlevels by a single factor experiment. The results showed that the optimum extractionconditions of Guiqi polysaccharide were as follows:solid-to-solvent ratio is1:10,extraction temperature of90℃,time contact of2h,and ratio of Angelica and Astragalusweight is1:5. According to the extraction method, extraction ratio of Guiqipolysaccharide is9.11%.To study the antioxidation of Guiqi polysaccharide, Angelica polysaccharide,Astragalus polysaccharide and Vitamin C, hydroxyl radical scavenging ability,superoxide anion radical scavenging ability, reducing power, and the inhibition of lipidperoxidation of them were determined and compared. The results indicated that Guiqipolysaccharides has higher and multiply antioxidant activity.Cylinder plate method and microdilution method were used to study antibacterialactivity of Guiqi polysaccharide against the six kinds of strains (E.coli, Bacilluslicheniformis, Streptococcus pneumoniae, Pseudomonas aeruginosa, Streptococcusagalactiae and Staphylococcus aureus). The results indicated that Guiqi polysaccharide has similar antibacterial activity against gram-negative bacterium or gram-positivebacterium by cylinder plate method and microdilution method.Method thiazolyl tetrazolium (MTT) was used to study anti-tumor activity ofdifferent concentrations of Guiqi polysaccharide against Hela, HepG2and A549cells invitro. The results showed that the different concentrations of Guiqi polysaccharide couldinhibit multiplication of cancer cells, and were dose-dependent with Guiqipolysaccharide concentrations (62.5μg/mL1000μg/mL) except31.25μg/mL group,have a significant difference as compared with the control group(p<0.01,p<0.05).Cell survival rate and inhibition rate were served as target to evaluate antiviralactivity of Guiqi polysaccharide against enterovirus71in vitro using method thiazolyltetrazolium (MTT) through Guiqi polysaccharide inhibit replication of enterovirus71,prevent enterovirus71from adsorbing and inactivate enterovirus71directly. Theexperiment results indicated that Guiqi polysaccharide prevent enterovirus71fromadsorbing vero cell was the most effective in these three modes of action.The anti-aging effort was found that Guiqi polysaccharides can obviously increasethe activation of GSH-PX, Hyp, and the XOD in liver tissue, and decrease the level ofH2O2, XOD and MAO of aging mice(p<0.01). The results proofed that Guiqipolysaccharides had the obvious effect on antisenility through the change of theseenzymes.The murine H22hepatocarcinoma served as an tumor model were used toresearch antitumor efficacy of Guiqi polysaccharide in vivo.From this experiment,Guiqi polysaccharide may play a major immunologic modification to show acertain antitumor efficacy for H22tumor-bearing mice.Carbon tetrachloride (CCL4) established acute liver impairment on mice model toestimate the protective efforts of Guiqi polysaccharides on liver impairment. The resultsshowed that Guiqi polysaccharide can significantly decrease the level of ALT and ASTin mice serum(p<0.01), liver tissue MDA content and liver index(p<0.01), increasedSOD activity(p<0.01), alleviate pathological changes in liver cell. The results indicatedthat Guiqi polysaccharide have obvious effect for protecting liver.
