Objectives: To explore the influence of electro-acupuncture at Yuan and Luo acupoints of liver and gall bladder meridians on the liver injury in rats with acute pancreatitis(AP)based on the theory of “combination of Yuan and Luo acupoints”;to explore the mechanism of alleviating gastrointestinal injury in rats with AP by electro-acupuncture at Shu and Mu acupoints of lung and large intestine meridians based on the theory of “exterior-interior relationship between the lung and the large intestine”.Methods: Part 1:All SD rats were divided into 3 groups(n=18): Normal Group,Model Group,Treatment Group(LR 3,GB 37).The rat model of AP was established by retrograde infusion of 3.5% sodium taurocholate into biliary pancreatic duct.TG received 3 times electro-acupuncture and 20 mins each time within 24 hours after establishing models.NG and MG just were kept rats at the same position with the same time as TG.The heart blood,pancreatic tissue and liver tissue were took 36 h after model establishment for the amylase(AMS),alkline phosphatase(AST),alanine aminotransferase(ALT),alkline phosphatase(AKP),tumor necrosis factor(TNF-α),interleukin 10(IL-10)test and pathologic observation.Part 2:All SD rats were divided into 4 groups(n=32): Group A(Da Chengqi Decoction(DCQD)),Group B(BL13,BL25,DCQD),Group C(LU1,ST25,DCQD)and Group D(BL13,BL25,LU1,ST25,DCQD).The rat model of AP was established by retrograde infusion of 3.5% sodium taurocholate into biliary pancreatic duct.All groups were offered with DCQD by intragastric infusion after 24 h of establishing models.Group B,C and D received 3 times electro-acupuncture and 40 min each time within 24 hours after establishing models.The heart blood,pancreatic tissue and intestinal tissue were took 36 h after model establishment for tumor necrosis factor(TNF-α),interleukin 4(IL-4),diamine oxidase(DAO),cholecystokinin(CCK),vasoactive intestinal plypeptide(VIP)test and pathologic observation.Results: Part 1: Compared with NG,the level of AMS,ALT,TNF-α were higher in MG(p<0.05),but the differences between two groups in level of AST,AKP and IL-10 were insignificant.Compared with MG,AST,ALT and TNF-α were significant higher in TG(p<0.05),but the differences between TG and MG in level of AMS,AKP and IL-10 were insignificant.Part 2:Compared with Group A,Group C had higher IL-4 and CCK level(p<0.05),and lower TNF-α level(p<0.05).The level of DAO in Group B and D(P<0.05)was better than that in Group A.The differences of VIP between Group B,C,D and Group A were insignificant;and VIP in Group C was lower than that in Group B and VIP in Group D was higher than that in Group C(P<0.05).Conclusion:Part 1: Three times electro-acupuncture at Tai Chong(LR 3)and Guang Ming(GB 37)in first 24 h after model establishment had some bad impact on the liver injury in rats with AP with unknown mechanism,which provide a clue for not recommendation the electro-acupuncture treatment for the liver injury complicated with AP.Part 2:The mechanism of electro-acupuncture at Shu and Mu acuacupoints for relieving gastrointestinal injury of AP rats is associated with inflammatory response reducing,gastrointestinal motility improving and gastrointestinal protection.Mu acupoints were better at inflammatory response;and Shu and Mu acupoints were better at gastrointestinal protection and gastrointestinal motility.
