| Objective: Based on the Interior-exterior relationship of the kidney and bladder meridians,to explore whether the electro-acupuncture(EA)at Yuan-primary acupoint of kidney meridian and Luo-connecting acupoint of bladder meridian could ameliorate the acute kidney injury(AKI)complicated with acute pancreatitis(AP)in rats and its mechanism.Methods: Eighteen male Sprague dawley(SD)rats were averagely divided into three groups at random,marked as the control group(group A),model group(group B),treatment group(group T)separately.Rats of acute pancreatitis in group B and group C were induced by retrograde perfusion 3.5% sodium taurocholate into the biliopancreatic duct,while the control group received the same volume of normal saline instead.Two hours after operation,rats were performed EA for the group T or fixed for the same time in every eight hours,lasting for thirty minutes each time in the group A and B as group T.Twelve hours after EA or fixed,all rats were sacrificed and the blood from heart,pancreas and kidney tissue sample were collected.Amylase,blood urea nitrogen(BUN),serum creatinine(SCr),Kidney injury molecule-1(KIM-1),Interleukin-8(IL-8)and Interleukin 10(IL-10)in serum were measured by enzyme linked immunoabsorbent assay(ELISA).pancreas and kidney tissue were used for pathological examinations.Results: the levels of amylase,BUN,SCr,KIM-1 and IL-8 in blood of rats in model group were significantly higher than those in the control group(P<0.05);there were no difference in IL-10 between model group and control group(P>0.05).The histopathological scores of kidney and pancreas in model group were significantly higher than those of control group(P<0.05).Compared with model group,the levels of amylase,SCr,KIM-1 and IL-8 in treatment group were decreased significantly(P<0.05)while IL-10 in treatment group increased significantly(P<0.05).The histopathological scores of both kidney and pancreas of rats in treatment group were obvious lower than those in the model group(P<0.05).Conclusions: EA at Yuan-primary acupoint of kidney meridian and Luo-connecting acupoint of bladder meridian could lower the pro-inflammation factors IL-8 and increase the anti-inflammatory factors IL-10 to balance inflammatory response and eventually ameliorate the acute kidney injury associated with acute pancreatitis. |
