Objective:Based on the early determination of the water-soluble alkaloids of Yimucao Injection is the effective material on the basis of its promote uterine contraction,the further study on the pharmacological basis and mechanism of the Yimucao Injection on effects of promoting uterine contraction and preventing hemorrhage.Methods:1.In vitro uterine smooth muscle activity test.YMCI-WTA was prepared by cation exchange resin purification method.Preparation of rat isolated uterine model and guinea pig isolated uterus,given the water-soluble total alkaloids0.05,0.1,0.2 mg/mL,with Powerlab sixteen channel physiological recording system records 10 min after uterine smooth muscle contraction curve,compared the changes of uterine contraction activity,frequency and tension.2.In vivo uterine smooth muscle activity test.Preparation of rat uterus in vivo model and the model of guinea pigs in the body of the uterus,intramuscular injection of YMCI-WT?10,20,40 mg/kg?,the BL-420 biological function experimental systeme records within 60 min dosing rat and guinea pig animals within 90 min of uterine smooth muscle contraction curve,uterine contractions were observed before and after comparison dosing activity,the change of frequency and tension.3.Experiment of Medical abortion guinea pigs.Mifepristone combined misoprostol medicine flow method is used to establish the guinea pig model of abortion,after the success of the building,the animal is divided into model group,YMCI-WTA group,to medicine group by gastric 0.17 g/kg of YMCI-WTA,7 days in a row,record the guinea pig uterus blood loss,after the last dose of blood ELISA method for determination of serum E₂,P content,determination of the proportion of uterus,and use the kit determination of NO and ET-1 in uterine tissue and calcium content.4.Promote uterine contraction active ingredients screening test.Using rat and guinea pig uterus in vitro model,give total water-soluble alkaloids of Yimucao injection Stachydrine,choline,trigonelline,using BL-420 biological function experimental system records within 10 minutes after the treatment of uterine smooth muscle contraction curve,uterine contractions were observed before and after comparison dosing activity,the change of frequency and tension;using guinea pigs in vivo uterine model,give total water-soluble alkaloids of Yimucao injection Stachydrine,choline,trigonelline,oxytocin and Yimucao injection,using BL-420biological function experimental system records within 90 minutes after the treatment of uterine smooth muscle contraction curve,uterine contractions were observed before and after comparison dosing activity,the change of frequency and tension;using the guinea pigs in vitro model of uterus,given different receptor blockers or agonist,then to observe the effects of Stachydrine on uterine activity.Results:1.Effect of isolated uterine smooth muscle activity:0.05 mg/mL of YMCI-WTA can be significantly enhanced the rat uterine contractions in vitro activity,the frequency and the maximum,0.1,0.2 mg/mL of uterine contraction,frequency,average and maximum increase trend;0.05,0.1,0.2 mg/mL of YMCI-WTA can obviously enhance the guinea pigs in vitro uterine contractions maximum.2.The impact on the in vivo uterine smooth muscle activity:10 and 40 mg/kg of YMCI-WT intramuscular injection can enhance the vitality of normal rats in vivo uterine contractions,frequency,average value;10,20,40 mg/kg of YMCI-WT can increase the normal guinea pigs in vivo uterine contractions maximum.3.The influence on guinea pig miscarriage uterus.0.17 g/kg of YMCI-WTA lavage abortion model guinea pigs,can reduce the bleeding,uterine significantly reduced proportion coefficient,and organization of the uterus NO and P content,increasethe uterus tissue calcium,ET content.4.The promote uterine contraction active ingredients screening results.In vitro uterine test,3×10^-5mol/L stachydrine,choline,fenugreek alone can significantly reduce the normal uterine contraction in rats the maximum,significantly increased guinea pig normal uterine contraction activity and reduce In the uterine test,intramuscular injection of 1mg/kg of stachydrine can significantly enhance the contraction activity,mean value and maximal value of the guinea pigs in the uterine uterus,1mg/kg of choline can significantly reduce the 60 to 80 minutes not pregnant guinea pig uterine contraction frequency,significantly increased after administration within 10 minutes uterine contraction maximum,1mg/kg fenugreekine can significantly increase after administration 20 to 40 minutes not pregnant guinea pig uterine contraction activity,20 to 90 minutes maximum contraction;0.83 U/kg oxytocin can significantly increase within 90 minutes after the treatment was not pregnant guinea pigs in vivo uterine contraction activity,average and maximum value,can significantly increase the dosage within 10 to 30 min after normal guinea pigs in vivo uterine contraction frequency,10 to 80 minutes minimum contraction,0.17mL/kg of Yimucao injection can significantly increase the drug delivery within 90min after normal guinea pigs in vivo uterine contraction force,average,maximum contraction within 80 min,contraction frequency within 10 to 20 minutes,10 to 90minutes contraction minimum value;L-type Ca²⁺channel blocker verapamil can significantly antagonize the stachyramine on isolated guinea pigs in vitro uterine excitatory effect.conclusions:1.YMCI-WTA can excite the normal guinea pig in vitro uterus and in vivo uterus,with the role of promoting uterine contraction,is the important pharmacodynamic material basis of Yimucao Injection on promoting uterine.2.YMCI-WTA can reduce miscarriage guinea pigs blood loss and promote restoration of the uterus,mechanism and regulation of uterine tissue vasomotor related material,may also be related to its promoting uterine contractions.3.Stachydrine,choline and fenugreek are the active ingredients of Yimucao Injection,and the action mechanism of the stachydrine may be related to the excitatory Ca²⁺channel.
