| Objective: To explore the possible mechanism of tonifying kidney compound in the treatment of thin endometrium,to provide a preliminary basis for the expansion of its application in the field of assisted reproductive diseases.We observed the effect of tonifying kidney compound on the expression of VEGF and Ang-2 in the thin endometrium of rats.Methods: In this study,we selected 80 female SD rats with 8-week-old SPF-grade sexual maturity without mating.Meanwhile,they had a weight of about 220~250 gram each rat.After one week of adapative feeding,as well as doing vaginal smears examination at 8 am each day to record the rat sexual information.Then we picked 75 rats out those who had two or more consecutive estrous cycles,and they were divided randomly into three groups:10 rats in the control group,10 rats in the sham operation group and the remaining 55 rats in the surgical group.The rats' uterine cavities in the operative group were treated with95 % alcohol-0.5 milliliter and retained 5 minutes.Three rats dead due to anaesthesia during the process of modeling.Next,we chose the successful model 52 rats,which were randomly divided into four groups: the model group,Kidney-reinforecing recipe low dose group,the middle dose group and the high dose group.The distributions of drugs each group just as followed:(1)The control and blank group: drinking water freely;(2)The model group: feed the rats with saline according to the dose 1ml/100g;(3)The kidney-reinforecing recipe small dose group: according to the dose 5g/kg?d by the free decoction granules;(4)The middle dose group: according to the dose 10g/kg?d by the free decoction granules;(5)The kidney-reinforecing recipe high dose group: 20g/kg?d crude drug by the free decoction granules;The frequency was one time each day,and orally for three estrous.After the last injection,as well as the rats were in the estrus cycle,we killed the rats and took the test samples.Results: 1.Compared with the blank group,the thickness of the endometrium in the model group was more thinner obviously,and the difference was statistically significant(P<0.01);Compared to the model group,the thickness of endometrium in the recipe groups had a noticeable increase,the difference between the recipe groups and the model group was statistically significant(P<0.01);2.When compared under an optical microscope,we can see that the endometrial epithelial cells integrity in the blank group were columnar and arranged regularly,and the quantity of the gland and blood vessel was normal.Meanwhile,the interstitial cells arranged evenly;When compared with the control group,the endometrium in the model group became thinner and arranged discontinuously,the epithelial cells of endometrium grew into low columnar and arranged irregularly.The quantity of the glands and blood vessels reduced obviously;After the medication,the thickness of endometrium rose along with its continuity.At the same time,it had an significant increase on the glands as well as the blood vessels;3.Compared with the model group,the expression of VEGF and Ang-2 was higher in the other groups,the difference was statistically significant(P<0.01),and the middle and high dose of Kidney-reinforcing recipe had significant statistical significance(P<0.01).Conclusion: The experimental study was to verify the academic thought of treating the thin endometrium by reinforcing kidney.2.Kidney-reinforcing recipe can promote the growth of the endometrium and increase the glands as well as blood vessels,and its mechanism may be mainly controlled by regulating the expression of vascular growth related factors VEGF and Ang-2. |
