Mango leaves are dried leaves of mango(Mangifera indica L.,Archichlamydeae,Sapindales,Anacardiaceae).The leaves as a traditional Chinese medicine,are cheap and easy to get and can treat the heat accumulation of abdominal pain and abdominal distension disease in clinic,decocting washing treatment of ulcer.Study on pharmacognostic identification of mango leaf medicinal materials in Guangxi and Yunnan Province.The identification of different varieties of mango leaves at base,the original plant morphology,properties,powder,different cross section.Provide the basis for the differentiation and further development of the medicinal materials.Mangiferin is the main active ingredient of mango leaves,which is widely used in clinic because of its non-toxic and no side effects.Recent studies have shown that mangiferin has antibacterial,anti-inflammatory,anti-diabetic,anti-tumor,anti-lipid metabolism abnormality and estrogen-stimulating effect.Combined with the etiology of ALD and the pharmacological activity of mangiferin,it is speculated that mangiferin may have therapeutic effect on ALD,which was also confirmed in our preliminary experiment.The ALD model,which is 40% alcohol induced,is similar to ALD caused by chronic excessive drinking,and is an ideal model for the study of ALD disease.This paper discusses the identification of mango leaves and the extraction and purification of mangiferin.And through the detection of liver function,the related biochemical indicators of measurement,histologic examination and application of real-time fluorescence quantitative PCR technology,it evaluates whether mangiferin has the efficacy and functions of anti-ALD from many aspects,and preliminarily discusses its related mechanism.The main research contents are as follows.1.The identification of mango leaves and the extraction and purification of mangiferin.There are great differences in leaf color,vessel,non glandular hairs,stone cells,calcium oxalate crystal and transverse section of leaves of different varieties,sometimes scattered on both sides of main veins,sometimes continuous semi ring.Thick wall tissue,secretory cell,medulla,medullary ray,oil chamber is much less.The relative sizes of the diameter of the primary xylem and secondary xylem are sometimes different,and sometimes they can be distinguished.The above conclusions can be used as the basis for the identification of mango leaves in different varieties.The content of mangiferin in mango leaves of different varieties was detected.We found that the mangiferin was the most abundant in the Baise mango leaves.Therefore,baise mango leaf was used as the material.After obtaining a series of treatments,such as ethanol extraction,petroleum ether extraction,decolorization,low pressure heating,concentration,filtration,recrystallization,concentration and filtration,the mangiferin crude product was obtained.Then the polyamide column chromatography was used to obtain the purified mangiferin from the gradient elution of water-ethanol system.The yield was 77.6%.Finally,the purity of mangiferin determined by HPLC was compared with the standard mangiferin.The purity of purified mangiferin was 98%,which is fully consistent with the requirements of the follow-up mangiferin pharmacology experiment.It provides a reference for industrial extraction and isolation of mangiferin.2.Protective effect of Mangiferin on alcoholic liver disease in ratsThe SD rats were randomly divided into normal group,model group,silibinin group(SYN,20 mg/kg)and MFN high-dose,low-dose groups(100?50 mg/kg).An ALD rat model was established by intragastric administration of 40% ethanol(10 m L/kg),medication was given at the same time.After 4 weeks of drug delivery,the concentrations of serum ALT and AST,TBIL,DBIL and IBIL were detected.The mRNA level of NF-?B and the contents of TNF-? and IL-6 in liver tissues were measured.Hematoxylin eosin(HE)staining and Oil red O staining were used to observe the pathological changes of liver tissue.Compared with normal group,the index of liver,spleen and kidney of rats in model group increased(P<0.01),the serum indexes of ALT,AST,TBIL,DBIL and IBIL were increased(P<0.01).NF-?B mRNA,TNF-? and IL-6 levels in liver tissue were increased(P<0.01).At the same time,a large number of vacuolar lesions,swelling of liver cells,fatty degeneration and inflammatory cell infiltration were observed in the liver.Compared with model group,the above-mentioned indexes in MFN high and low dose groups were significantly improved(P<0.01,P<0.05),and the degree of liver cell lesion was obviously alleviated and dose dependent.In summary,there is a significant difference between different varieties of mango leaf in the identification of raw drugs,and its pharmacological identification is the scientific basis for the differentiation and further development of the medicinal herbs.In this experiment,the extraction and purification of mangiferin can completely accords with the requirements of the follow-up mangiferin pharmacology experiment.Mangiferin has a good anti-ALD effects,the mechanism of action is related to hepatocyte repair,regulation of liver cell inflammation(especially NF-?B,TNF-?,IL-6 levels).
