ObjectiveAquilaria is the wood of Aquilaria sinensis(Lour.)Gilg,has a very high commercial and medicinal value in the world.As one of Chinese important economic crops,Aquilaria sinensis has massive planting area including Guangdong,Guangxi,Yunnan,Hainan and other place.The periodism of the leaf of Aquilaria sinensis is twice a year,which have great output and always been fired and abandoned,which is great waste;This paper studied the extraction and purification process of the leaf of Aquilaria sinensis:optimization of mangiferin ultrasonic extracting process through the Response Surface Method(RSM),The concentration of mangiferin of the extractive of Aquilaria sinensis was purified by macroporous resin.The study aimed to decrease the waste of the leaf and increase the sources of mangiferin.Compared the antioxidant capacity among Mangiferin,Vitamin C and BHT,and provide the basis for the research and development of industrial and research on the development of food antioxidants.Methods1.Measure the content of mangiferin in the leaves of Aquilaria sinensis by methodology.Through single factor experiment investigate the best position:ultrasonic temperature,ultrasonic extraction time,ethanol concentration,ultrasonic power and solid-liquid ratio on the extracting yield,Selecting three factors:the extraction time,the solid-liquid and the concentration of ethanol were used to simulate the Box-Behnken design experiment,and the mathematical model was developed by response surface method to predict the best extraction process.2.Macroporous resin purified the contents of mangiferin in the leaf of Aquilaria sinensis,through static adsorption and desorption of six kinds of macroporous resin,buliding static adsorption-desorption kinetics curve of macroporous resin,determine the loading flow rate of the sample and the height of the column and the concentration of alcohol,Verified the process by the experimental.3.Antioxidant experiment in vitro by scavenging DPPH· radicals,ABTS+·radicals,Cu2+ reduce ability,comparing the ability of antioxidant among mangiferin,VC,BHT.ResultsThe results of Single factor experiment:ultrasonic extraction time is 30 min,ultrasonic frequency is 100 kHz,solid-liquid ratio is 15:1,temperature of 35? and 50%ethanol.By the response surface method optimization process with 58%ethanol,solid-liquid of 16.1:1,extract time of 39 min,predict the response value is 1.78%,the regression coefficient R~2 is 0.9577,adjustment coefficient Radj2 is 0.9032,the actual values is 1.78%;There were significant differences in the desorption and adsorption of mangiferin of five macroporous resins with AB-8(P<0.05),The adsorption time of mangiferin was 120 min,the desorption time was 60 min?The amount of adsorption of the AB-8 was 2.20g/mg,when the sample flow rate is 2 ml/min?The high is 25 cm?The elution concentration is 30%.After froen by crystallization,the purity was 80%mangiferin;By antioxidant experiment in vitro,mangiferin,VC and butylated hydroxytoluene' s IC50 of eliminate ABTS+· were 51.12 ±2.72,36.58 ±1.70 and 354.88±1.27 ?g/mL,IC50 of eliminate DPPH· were 125.43±7.52,91.72+5.35,768.19±11 30 ?g/mL,IC50 of Cu2+ reduction ability were 73.27± 0.92,327±2.18,349.31±2.89 ?g/mL.ConclusionThe regression curve R~2 =0.9577 and The adjust curve Radj2 =0.9032,which indicates that the experimental design method can explain 90.32%of the model.The actual value measured by the simulation experiment is close to the predicted value,The optimized extraction process of mangiferin was feasible and reliable.After purification by macroporous resin,the content of mangiferin was increased by 50%,and the purity of 80%mangiferin was obtained by frozen crystallization.Therefore,the optimum extraction was optimized by the response surface method The results showed that the higher the concentration of mangiferin,the stronger the antioxidant activity,the scavenging rate of DPPH· and ABTS+· were 72%and 73%of VC,While the copper ion reduction ability is stronger than VC,the antioxidant capacity is stronger than BHT,and no toxicological experiment report.so mangiferin can be used as natural antioxidant development research.
