Objective:Buzhong Yiqi-Pill is a famous prescrition about replenishing energy and blood,which is widely used in clinic.In recent years,it is reported that it serves as the supplementary therapy of bone marrow suppression after chemotherapy.It can make hematological indexes and systemic symptom better.Bone marrow suppression is one of the common adverse reactions after chemotherapy.Peripheral blood cells are reduced by acute injury of bone marrow after chemotherapy,especially white blood cells,followed by platelets and red blood cells.Severe cases can reduce hemoglobin or even whole blood cells.This experiment will be designed to study about the regulation and mechanisms of Buzhong Yiqi-Pill on bone marrow hematopoietic system in order to provide laboratory data for later clinical use.Methods:bone marrow inhibition of mouse model will be established to simulate blood deficiency syndrome in the experimental subject.The study is going to use the experimental technologies of peripheral blood cell count,bone marrow pathological section,Semi-solid agar culture assay of hematopoietic progenitor cells in vitro,flow cytometry,etc.It will use peripheral blood count,bone marrow nucleated cell count,bone marrow morphology,hematopoietic progenitor cell survival and colony forming rate to study the effect of Buzhong Yiqi-Pill on bone marrow hematopoietic function of bone marrow suppression mice and explore its mechanism to provide experimental data support for subsequent drug application.Results:1.After giving Buzhong Yiqi-Pill,it can significantly increase the number of white blood cells,red blood cells,platelets and bone marrow nucleated cell in bone marrow inhibition of mouse mode.The middle dose group has the best effect among them.2.After giving Buzhong Yiqi-Pill,the area of bone marrow hematopoiesis of mice is restored to some extent and the hematopoiesis tissue area is increased.3.After giving Buzhong Yiqi-Pill,it is beneficial to the formation of Semi-solid agar culture assay of hematopoietic progenitor cells in vitro,in myelosuppressed mice including the formation of erythroid,granulocyte and megakaryocyte colony in vitro.Each cell colony has its own characteristics.4.After giving Buzhong Yiqi-Pill,it can prevent the apoptosis of bone marrow suppression in mice hematopoietic stem cell and hematopoietic progenitor cell.And it adds proliferation index?PI?of bone marrow cell and promote bone marrow cell to enter S phase and G2/M phase from quiescent G0/G1 phase.This will lead to the normal differentiation of bone marrow hematopoietic cells.Conclusion:Giving Buzhong Yiqi-Pill to mice with bone marrow suppression could repair their hematopoietic function after chemotherapy and make bone marrow hematopoiesis active.It is beneficial to increasing bone marrow suppression mice's the number of nucleated cells,peripheral blood cells andthe hematopoiesis tissue area.It also stimulate the formation of external agar culture of hematopoietic progenitor cells,including erythroid,granulocyte and megakaryocyte colony.It can also inhibit the programmed cell death of bone marrow cells andpromote bone marrow cells into the proliferation cycle.And then it let cells go out of dormancy phase,transforming G0/G1 phase into S phase andG2/M phase and completing normal cell cycle.Buzhong Yiqi-Pill regulateshematopoietic system in bone marrow suppression mice and restore its hematopoietic functionregulatemainly through the above methods.
