Objective:Chemotherapy is the main treatment of malignant tumor.But it has serious side effects,bone marrow inhibition is one of the more intractable side effects.The main manifestations of the patient's peripheral blood leukocytes,red blood cells,platelets and even hemoglobin levels decreased,even can cause whole blood cells to decrease in severe time.It hinders the treatment of the disease,affect the patient's basic life?However,the therapeutic effect of traditional Chinese medicine on bone marrow inhibition has been shown to be significant and the toxicity is small.But compared to previous experiments,mostly directly from the formula with tonifying blood to start.Therefore,this experiment is based on the herbal medicine of the Guishao Dihuang-pill,to understand the formula and the"tonifying bone marrow and blood"thinking of the treatment of the bone marrow inhibition and the regulation of hematopoietic system and the mechanism of the exploration.To obtaining experimental data and providing more ideas and options for clinical treatment.Methods:In this experiment,mice were used as a model object for bone marrow inhibition after chemotherapy.We applicate automatic blood cell analysis,pathological slices of bone marrow tissue,semi-solid agar culture technique in vitro of hematopoietic progenitor cells,and flow cytometry and other experimental methods.We count from peripheral blood cells,bone marrow nucleated Cell count,morphological manifestations of bone marrow tissue,survival of hematopoietic progenitor cells,and the rate of cell colony yield,bone marrow cell apoptosis and proliferation cycle.Level and systematic study on the effect and mechanism of the bone marrow hematopoietic system function of bone marrow-inhibiting mice by the combination of nourishing bone,marrow and enriching the blood.Provide experimental support and more choices for clinical treatment of bone marrow inhibition?Results:1.After treatment with Guishao Dihuang-pill,a significant increase in the number of peripheral blood and bone marrow nucleated cells in mice with bone marrow inhibition.The medium dose group had better effect.2.After treatment with Guishao Dihuang-pill,can increase bone marrow inhibition of bone marrow hematopoietic tissue area in mice.3.After treatment with Guishao Dihuang-pill,the formation number of the three-system?red,kernel,and giant nuclei?in the mice with agar-semi-solid culture in vitro was increased and each set fell each.4.After treatment with Guishao Dihuang-pill,rduce the number of bone marrow cells in the G0/G1 at rest period and increase into the s and G2/M,and raise the PI exponent,bone marrow hematopoietic cells enter normal cell proliferation cycle and inhibit cell apoptosis.Conclusion:The hematopoietic function of mice with bone marrow inhibition caused by chemotherapy-induced hematopoietic system can be restored by giving the medicine to GuiShao Dihuang-pill.It can increase bone marrow inhibition of peripheral blood cells and bone marrow nucleated cells in mice,enlarged bone marrow hematopoietic tissue area,stimulation of agar culture in vitro for the generation of hematopoietic progenitor cells including red,granulocyte and giant nuclei,and awaken dormant bone marrow cells.To start the multiplication cycle and enter the normal reproductive cycle of the G0/G1 stage in the S period?G2/M period,so as to inhibit the apoptosis of bone marrow cells.The Guishao Dihuang-pill mainly participates in the regulation of hematopoietic function by bone marrow inhibition in mice through the above-mentioned pathway.
