Objective: Useing the last 15 days of chronic restraint combined chronic pain stimulation to duplicate insomnia rat model with syndrome of liver depression transforming into fire.Test the model from three aspects: macroscopic characterization,biological parameters and Syndrome Detecting from Recipe Used.Methods: Dividing the 30 healthy SD rats to blank group,model group and treatment group,with each group of 10 rats.The rats in the model group and the treatment group are treated with chronic restraint(restri-cting activity)combined with chronic pain stimulation(tails with forceps clip)for 15 days.After the modeling and its behavioral tests,the treatment group was treated with Long dan xie gan Decoction and zhu sha an shen wan continuous intragastric administration for 5 days,at the same time,the model group wastreated with equal pure water.Observe the general condition,food intake and body weight of rats,the spontaneous activity of rats was measured for 24 hours,the elevated cross maze test,open field test,tail suspension test,MD of LC-NE and the amount of plasma NE.Results:(1)The general condition: before modeling the rats in each group had a good sleep,normal activities,quick-minded,iustrous eyes,eye socket and lips are pink,claw smooth,bright white fur,psychosis is good,when grasping,resistance is fierce when grasping.After 5 days of continuous modeling,the model rats began to appear nervous,sensitive to the outside world,the hair was slightly messy,bound to resist violent but still flexible reaction;After 10 days of continuous modeling,the attack of model rats increased,irritability,struggle in bondage,the ear tip slightly dry coke,and the nail tip mucosa color dark red.After 15 days modeling,the spirit of model rats of high tension,a sound into defensive stance,anxious eyes,messy hair,lack of luster,biting each other and a significant increase in the number of attacks,bound hammer grab violent struggle,screech,aggressive,ear contour of dry coke,nasal mucosa and limbs fingertip redness slightly loose stool.(2)The increase of weight: After 5 days of continuous modeling,there is no statistic difference(P>0.05)in weight gain.After 10 days of continuous modeling,the weight gain of the model rat is significantly reduced within the last 5 days(P < 0.01),There is no statistic difference between the model group and the treatment group(P>0.05).After 15 days modeling,the weight gain of the model rat is increased significantly reduced within 5 days.(P<0.01).There is no statistic difference between the model group and the treatment group(P>0.05).After treatment,the weight gain of rats in the model group was significantly reduced during the 5 day treatment(P< 0.01),the weight gain of the treatment group in the 5 day treatment period was significantly improved(P < 0.01),close to the blank group(P> 0.05).(3)Food intake: After 5 days of continuous modeling,the rats intake has no significant difference(P > 0.05).After 10 days of continuous modeling.the food intake of model rats significantly reduced(P < 0.01),there is no significant difference between the model group and treatment group(P> 0.05);After 15 days of continuous modeling,the food intake of model rats decreased significantly(P < 0.01),there is no significant difference between the model group and treatment group(P> 0.05);after the treatment,The food intake of model group was significantly decreased(P< 0.01).the rats in the treatment group increased significantly(P < 0.01),close to the blank group(P > 0.05).(4)24 hours of spontaneous activity : After 15 days of continuous modeling,the daytime activity of model rats increased(P < 0.01),there is no significant difference between the model group and treatment group(P > 0.05);the night activity of model rats slightly decreased,but no significant difference between the groups(P > 0.05),after the treatment,the daytime activity of rats in the model group increased(P < 0.01).the night activities of model rats decreased(P > 0.05).The daytime activity of treatment group rats were significantly decreased(P < 0.05),close to the level of the normal group(P > 0.05);the activity of the rats in the treatment group increased at night(P > 0.05).(5)The elevated plus maze test: After 15 days of continuous modeling,the OE% and OT% values of model rats decreased significantly(OE% P < 0.01;OT%: P < 0.05),there is no statistical significance difference between the model group and treatment group(P > 0.05);after the treatment,the OE% and OT% values of model group rats decreased significantly(OE%:P < 0.01;OT%:P < 0.05).the OE% and OT% values in the elevated plus maze test of the treatment group were significantly higher(OP< 0.05),which was close to the normal group(P > 0.05).(6)Open field test: After 15 days of continuous modeling,The open field experiment in central square score,vertical scores,modification times and feces of model rats significantly increased(P < 0.05),there is no significant difference between the model group and treatment group(P > 0.05).After the treatment,the model group rats in the open field experiment in central square score,vertical scores,modification times and feces increased obviously(P < 0.05),the treatmeat group rats in the open field experiment in central square score,vertical scores,modification times and feces decreased obviously(P < 0.05).(7)Rat tail suspension test: After 15 days of continuous modeling,the rat tail suspension immobility time of model rat is increasing,but there was no significant difference between the three groups of rats(P > 0.05);after the treatment,there was no significant difference between the three group in mouse tail suspension immobility time(P > 0.05).(8)Plasma NE content: compared with the blank group,the content of NE in plasma of the rats in the model group increased significantly(P < 0.01);compared with the model group,treatment group,plasma NE content of rats decreased significantly(P < 0.01),close to the level of the blank control group(P > 0.05).(9)MD of LC-NE: compared with the control group,MD of LC-NET in rats of model group significantly increased(P < 0.01);compared with the model group,the treatment of MD of LC-NET in rats significantly decreased(P=0.010 < 0.05),close to the level of the blank control group(P > 0.05).Conclusion:(1)The macroscopic evaluation and biological evaluation showed that the chronic restraint(restri-cting activity)combined with chronic pain stimulation(tails with forceps clip)modeling method could be used to duplicate insomnia rat model with syndrome of liver depre-ssiontransforming into fire.(2)Syndrome Detecting from Recipe Used of Longdanxiegan Tang combined zhushaanshenwan gavage treatment show that model of chronic restraint(restri-cting activity)combined with chronic pain stimulation(tails with forceps clip)could be used to duplicate insomnia rat model with syndrome of liver depre-ssiontransforming into fire.
