?Objective?:This study is based on "fine-blood-marrow one theory",That is,blood can nourish the kidney essence,to refine the marrow,but also directly into the brain to replenish the brain,the blood has the dual role of nutrient brain.The experimental study selected vascular dementia animal brain hippocampal CA1 region as the observation area,with astrocyte morphology,quantity changes and changes in microvascular neovascularization density in the ischemic area as the specific observational indicators,to study the relationship between learning and memory abilities,brain histomorphology,number of GFPA-expressing immunoreactive cells,and microvessel neovascularization in different doses of Dangguibuxue Decoction in VD rats,to explain the modern biological basis of Danggui Buxue Decoction in treating vascular dementia,and find the best dosage of Danggui Buxue Decoction in treating vascular dementia and to further promote the theory of "sfine-blood-marrow one theory" in clinical application to find theoretical and scientific support.?Method?:Selection of young male SD rats as experimental subjects.Modeling Vascular Dementia with Modified Pulsinelli Four Vessel Blocking(4-vo).Seven days after operation,the behavior of the model rats was observed and scored according to the degree of neurological deficits.Those who failed to make models and had excessive nerve damage were excluded.the rest were selected to enter the experiment.The experiments were divided into normal group,sham group,VD model group,LDG group,MDG group,HDG group.According to the experimental need to use different drugs and doses,once a day,a total of intervention for 90 days,and then Morris water maze test to detect changes in spatial cognitive ability of each group of rats.After water maze,the brain tissue of each group was extracted.Hematoxylin-eosin(HE)staining was used to observe the brain histomorphology under light microscope;immunohistochemical staining of vascular endothelial cell factor VIII factor was performed to observe the density of microvessels around the ischemic brain tissue of brain tissue;The number of immunoreactive cells expressing GFAP(collagen fibrillary acidic protein)in the hippocampus of rats in each group was examined by techniques.To compare the effects of Dang Gui Bu Xue Decoction on the degree of changes of indicators in different doses and to analyze the reasons of Dang Gui Bu Xue Decoction improving the ability of learning and memory in VD rats.?Result?: 1.Morris water maze behavior test results:The water maze escapes latency,differences between the VD,VD model + Danggui Buxue Decoction group and the sham operation group were significantly longer(P<0.01);there was no difference between the normal group and the sham operation group(P>0.01);All VD models + Angelica Biloba Compared with the VD model group,the group was significantly shorter(P<0.01);VD model + high dose Danggui Buxue Decoction group compared with the VD + middle dose,VD + low dose group,the time was shortened,the difference was significant(P <0.01)2.Morphological observations : The morphology and distribution of neurons in hippocampal CA1 region in normal and sham-operated groups were normal,with almost no degeneration and deterioration.Dead neurons.In the VD model group and in the doses of Danggui Buxue Decoction group,neurons and apoptotic neurons were observed in the CA1 area.3.GFAP expression immuno-positive cell count results:Compared with normal group and sham operation group,the number of GFAP expression in VD model group,VD model + each dose of Danggui Buxue Decoction group was significantly higher(P<0.01).There was no significant difference between VD model group and VD model + low dose Danggui Buxue Decoction group(P>0.01).VD model group,VD model + middle dose of Danggui Buxue Decoction group,VD model + high dose Danggui Buxue Decoction group,the number of GFAP expression increased in turn,and there were significant differences among groups(P<0.01).There was no significant difference between the normal group and the sham group(P>0.01).4.Ischemic brain microvessel density test results:Compared with normal group and sham operation group,VD model group,VD model + Danggui Buxue Decoction group had higher microvessel density,with significant difference(P<0.01).There was no significant difference in microvessel density between VD model group and VD model + low dose Danggui Buxue Tang group(P>0.01).VD model group,VD model + middle dose of Danggui Buxue Decoction group,VD model + high dose of Danggui Buxue Decoction group,and the density of microvessels in ischemic lesions and peripheral tissues increased sequentially,with significant differences among groups(P<0.01).There was no significant difference between the normal group and the sham group(P>0.01).
