Objective:To observe the effect of Electroacupuncture on apoptosis and ERK1/2 protein expression of hippocampal CA1 area in rats with vascular dementia(VD).Through the mechanism of electroacupuncture treatment in the frontal area of VD.The electro acupuncture treatment scheme was optimized to provide experimental basis for clinical treatment of VDMethods:73 Wistar rats were randomly selected,56 of them were randomly selected to make VD model,and the modified 2-VO method was used to make the model.The rats successfully made VD model were randomly assigned into the Electro acupuncture Group,Western Medicine Group and Model Group,with 15 rats in each group.The other 17 rats only underwent the blunt dissection of vagus nerve without clipping or ligation of common carotid artery.15 rats were randomly selected into the Sham Operation Group.The Electroacupuncture Group was selected according to the Yu professor's head partition method.Cluster needling from Shenting to Xinhui,and parallel lines 1 inch and 2 inches to the left and right.The frequency is 1-2HZ,treatment for 20min,and the needle is kept for 10min after the treatment.Once a day for 28 days.The Western Medicine Group was given piracetam solution(40mg/ml)once a day.The Sham Operation Group and the Model Group were grabbed and fixed once a day for 30 minutes without any treatment.After 28 days of treatment,Morris water maze test was used to detect the behavior of rats in each group.After the test,the hippocampus of the rats was taken.HE staining was used to observe the histomorphological changes of hippocampal cells in each group.The expression of Bcl-2,Bax and ERK1/2 proteins in the hippocampal CA1 area of rats and the changes of Bcl-2/B ax ratio were observed by immunohistochemistry.Electroacupuncture can inhibit apoptosis and regulate synaptic plasticity of nerve cellsResults:The results of HE staining showed that in arrangement of nerve cells in CA1 area of hippocampus in Model Group was disordered and not close,the cell structure was incomplete,the nucleus was pyknosis,fragmentation and dissolution.The nerve cells in the hippocampus of the Electroacupuncture Group were closely arranged.The cell structure is more obvious,clear nuclei can be seen,and the number of pyknosis and fragmentation nuclei is less.The results of immunohistochemistry showed that the ability of memory and learning of rats in the Model Group decreased significantly.The expression of Bcl-2 protein decreased,the expression of Bax protein increased,the ratio of Bcl-2/B ax increased significantly,and the expression of ERK1/2 protein decreased in the CA1 region of hippocampus in the water maze experiment(P<0.01).Compared with the Model Group,the escape latency and quadrant crossing times of Electroacupuncture Group were significantly increased,the expression of Bcl-2 protein in CA1 area of hippocampus was significantly increased,the expression of Bax protein was significantly decreased,the ratio of Bcl-2/B ax was significantly increased,and the expression of ERK12 protein was significantly increased(P<0.01).The Bcl-2 protein positive cells and the ratio of Bcl-2/Bax in the CA1 area of hippocampus in the Electro acupuncture Group were higher than those in the Western Medicine Group,and the Bax protein positive cells were lower than those in the Western Medicine Group.ERK1/2 protein positive cells in CA1 area of hippocampus in Electro acupuncture Group were significantly higher than those in Western Medicine Group(P<0.05).Electroacupuncture Group had better effect than Western Medicine Group,and had better inhibitory effect on cell apoptosis and regulation of synaptic plasticityConclusion:The treatment of electro acupuncture in frontal area can promote the expression of Bcl-2 protein,inhibit the expression of Bax protein,Bcl-2/Bax ratio increased and improve the expression of ERK1/2 protein in CA1 area of hippocampus of VD rats.Electro acupuncture at frontal area can inhibit apoptosis,regulate synaptic plasticity of nerve cells and protect neurons.Thus,the spatial memory and learning ability of VD rats were improved,and the curative effect was better than that of Western medicine.
