?ObjectiveThis study is based on the theory of“ essence-blood-brain integrated ”,that on the one hand,blood can directly nourish the brain,and on the other hand the essence-blood-brian can be interbred,so blood has a dual function of nutrition of the brain.Therefore,putting forward by danggui buxue tang of invigorating blood in the treatment of vascular dementia(VD).Clinical finding of VD patients with poor compliance is result in bad curative effect,and under the guidence of this theory,preliminary research found that the invigorating blood in the treatment of VD is superior to the traditional invigorating kidney.The effect of high dose was the best for VD rats,so pick the best dose and time window of concentration-response is necessary to improve the curative effect,and observe the best point of danggui buxue tang to improve the perfusion of VD rat hippocampal CA1 area,promoting angiogenesis,inhibiting the relationship between the aging of neuron apoptosis,and providing scientific support for danggui buxue tang treatment vascular dementia,further promoting the theory of “essence-blood-brain integrated”application in clinical.?MethodIn healthy SPF SD male rats as experimental subjects,100 rats were randomly divided into VD model + high-dose angelica blood tonic soup 90 days group,high dose of angelica blood tonic soup + VD model 180 days + high dose group,VD model angelica blood tonic soup three groups for 270 days,the first two group of 33,34 only the third group.Modified Pulsinelli four vessels day age 22 block(4-VO)production of VD rats model,building a week after the behavior according to the model and neural function defect scale selected 49 only model test in rats,respectively is 15,16,18.After 7 days of mold making,the angelica decoction was given to the angelica granule,which was administered to the stomach for 90 days,180 days,and 270 days respectively.Using Morris water maze,immunofluorescence,HEstaining,light microscopy and immunohistochemical methodsafter 270 days,to observe the behaviorof VD rats,the number of hippocampal CA1 GFAP,factor ?,changes of morphology and brain microvessel density.?Results1.Morphology: the VD model + high-dose angelica blood soup group(3 groups)showed more hyperplasia of astrocytes in the hippocampus than in the model group,indicating the phenomenon of neurophagia,and 270 days < 180 days <90days group(two comparison has significant difference,p < 0.01).2.Morris water maze behavior test results: Morris water maze experiment to escape the incubation period of time,high doses of danggui buxue three groups escape latency time are shortened,and 270 days <180 days < 90 days group(two comparison has significant difference,p < 0.01).3.GFAP expression was observed under Fluorescence microscope Results: VD model high dose of danggui buxue 3 groups of rat hippocampal CA1 area of GFAP expression immunofluorescence cells were significantly apparent,and 270 days < 180 days <90days group(two comparison has significant difference,p < 0.01).4.Ischemic brain microvessel density test results:VD model +high dose of danggui buxue high dose group of 90 days,the VD model+ danggui buxue180 days group,VD model + high dose danggui buxue 270 days group,the density of the microvessels around the ischemia increased in turn.(two comparison has significant difference,p < 0.01).?Conclusion1.The high dose of danggui buxue can improve the learning and memory abiity of vascular dementia rats.2.The high dose of danggui buxue improve the VD rars learning and memory abiity was best at 270 days.3.The mechanism of best point of danggui buxue improve vascular dementia ratslearning and memory ability: regulated astrocytes in hippocampal CA1,promote angiogenesis,nutrition and repair damaged neurons.
