Objective:To study the mechanism of the improvement of idiopathic pulmonary fibrosis by Buyanghuanwu decoction,the idiopathic pulmonary fibrosis model of SD rats was reconstructed with one-time tracheal injection of bleomycin improvement mechanism of pulmonary fibrosis by using Buyanghuanwu decoction by injection of intravenous injection of bleomycin SD rats idiopathic pulmonary fibrosis model.To observe the changes of fibrosis,autophagy and apoptosis related factors,to find out the internal relation of the three,and to provide new ideas for the rational and effective application of the corresponding TCM treatment to the idiopathic pulmonary fibrosis.Methods:72 SD rats were divided into 6 groups at random: the blank control group,model group,positive control group and Buyanghuanwu decoction high dose group,Buyanghuanwu decoction medium dose group,Buyanghuanwu decoction low dose group,each group of 12.In addition to the blank control group,a disposable tracheal injection was used to make rats' pulmonary fibrosis model.The blank group was injected with the same amount of NS.Rats in each group were given intragastric administration on the second day after modeling.Different doses of drug were given to the high,middle,and low dose groups of Buyanghuanwu Decoction.The positive control group was given prednisone.The blank control group and the model group were given the same amount of pure water,once a day.Rats were killed and lung tissue collected after 28 days of gavage.HE staining and Masson staining were used to observe the pathological changes of lung tissue sections;Immunohistochemical staining was used to observe the expression of COL-I,Beclin 1,BCL 2,and Caspase3 in lung tissue;The LC3?,and p-BCL-2 were detected by western blot.SPSS20.0statistical analysis was used to obtain the results.Results:(1)Pathological changes of lung tissue: there were no obvious pathological changes in the blank control group.The pulmonary interstitial fibrosis was evident inthe model group,and all treatment groups were alleviated.(2)The expression of COL-?,Beclin 1,BCL 2 and Caspase 3:(1)COL-?:Compared with the blank control group,the expression of Col-I in lung tissue of the model group was significantly increased(P<0.01).Compared with the model group,the expression of COL-? in the lung tissue of the positive control group was decreased(P<0.01).COL-?expression in the lungs of rats in each dose group of Buyanghuanwu Decoction decreased(P<0.01),and the highest dose of Buyanghuanwu Decoction decreased most significantly.(2)Beclin-1: compared with the blank control group,the expression of beclin-1 in the lung tissue of the model group was significantly reduced(P < 0.01).In comparison with the model group,the expression of beclin-1 in the lung tissues of the rats in the positive control group and the Buyanghuanwu decoction groups increased(P < 0.01).(3)BCL-2: compared with the blank control group,the expression of BCL-2 in the lung tissue of the model group was significantly increased(P < 0.01).In comparison with the model group,the expression of BCL-2 in the lung tissues of the rats in the positive control group and the Buyanghuanwu decoction group was decreased(P < 0.01).(4)Caspase-3:compared with the blank control group,the expression of caspase-3 in the lung tissue of the model group was decreased(P < 0.05).Compared with the model group,the expression of Caspase-3 in the lung tissues of the rats in the positive control group and the Buyanghuanwu decoction group was increased(P < 0.01).(3)The expression of LC3?,and p-BCl-2:(1)LC3-?:Compared with the blank control group,the expression of LC3? in the lung tissue of the model group was significantly decreased(P<0.01).Compared with the model group,the expression of LC3? in the lung tissue of the positive control group and the Buyanghuanwu Decoction groups(P<0.01).(2)p-BCL-2: compared with the blank control group,the expression of p-BCL-2 in the lung tissues of the model group was significantly increased(P< 0.01).Compared with the model group,the expression of p-BCL-2 in the lung tissues of the rats in the positive control group and the Buyanghuanwu decoction group were all decreased(P < 0.01).Conclusion :(1)A disposable tracheal injection of bleomycin can be used to reconstruct the idiopathic pulmonary fibrosis model of SD rats.(2)Buyanghuanwu Decoction can reduce the content of fibrogenic factors COL? in lung tissue,thereby improving pulmonary fibrosis.This may be one of the mechanisms of Buyanghuanwu Decoction against idiopathic pulmonary fibrosis.(3)BCL2 and p-BCL-2 are both of the interactive proteins of apoptosis and autophagy.Buyanghuanwu decoction can inhibit the expression of apoptosis factors BCL2 and p-BCL-2 and promote the expression of Beclin1,LC3? and Caspases3,thereby promoting the occurrence of pulmonary fibrosis apoptosis and autophagy.Combined with(2),it can be concluded that Buyanghuanwu Decoction may treat idiopathic pulmonary fibrosis by promoting apoptosis and autophagy in idiopathic pulmonary fibrosis tissue.
