ObjectiveTo apply bleomycin to iuduce the pulmonary fibrosis model on rats,through testing the content of HMGB1,?-SMA in lung tissue and IL-1?,IL-6,TNF-? in bronchoalveolar lavage of lab,to explore the possible mechanism of Buyanghuanwu decoction on pulmonary fibrosis and lay basis for clinical theatment.Method144 SD male rats were divided into 6 groups randomly:normal saline group? modeling group?prednisone group?high-dose Buyanghuanwu decoction group? middle-dose Buyanghuanwu decoction group and low-dose Buyanghuanwu decoction group,24 for each group.Firstly,except normal saline group,other groups were given 5mg/ml bleomycin into the trachea to set up pulmonary fibrosis model,at the same time,normal saline group given with equal volume of saline.Secondly,on the third day after the models were successfully made,each groups were treated with medication accordingly,continuous dosing 28 days,then specimen were taken three times each on the 7th?14th?28th day.Finally, Test the expression of HMGB1,?-SMA in lung tissue by immunohistochemical method and collect alveolar lavage to detect the content of IL-1?,IL-6,TNF-a with ELISA method.Result1?Compared with the normal saline group,the contents of HMGB1,a-SMA in lung tissue and IL-1?,IL-6,TNF-a in alveolar lavage fluid of model group were obviously increased(p<0.05);2?Each medicated group had different decreases in the contents of HMGB1,?-SMA in lung tissue and IL-1?,IL-6,TNF-a in alveolar lavage fluid of by comparion with model group(p<0.05);3?Compared with the prednisone group,the contents of a-SMA of Buyanghuanwu decoction low-dose group obviously decreased on the 28th day(p<0.05),as well as the level of TNF-a Buyanghuanwu decoction middle and low-dose group on the 7th,14th,28th day(p<0.05),while the expression of HMGB1,IL-1?,IL-6 showed an increase tendency,with a statistical significance,but still lower than model groups.Conclusion1?The PF rat model induced by bleomycin was successfully duplicated.2?Buyanghuanwu decoction could effectively control the content of HMGB1,?-SMA in lung tissue.3?Buyanghuanwu decoction could obviously decrease the level of IL-1?,IL-6,TNF-a in alveolar lavage fluid of experimental rats.4, It was improved that therapeutic effect of Buyanghuanwu decoction in PF were possibly associated with regulating the expression of HMGB1/?-SMA and its related factors in inflammatory state of PF.
