Research On The Effects Of Buyanghuanwu Decoction On The Expressions Of HMGB1,IL-8,α-SMA In Bronchoalveolar Lavage Fluid Of Rats With Pulmonary Fibrosis

Objective:The study was designed to observe influence effects of Buyanghuanwu decoction on histopathology of lung and the expressions of HMGB1, IL-8,a-SMA in bronchoalveolar lavage fluid from rats with bleomycin (BLM) induced pulmonary fibrosis to explore the efficacy and mechanism of Buyanghuanwu decoction in the treatment of pulmonary fibrosis.Methods:Sixty-four SD rats were randomly divided into four groups as sham operation group, model group, prednisone group and Buyanghuanwu decoction group. Model group, prednisone group and Buyanghuanwu decoction group were intratracheally administered with bleomycin to produce pulmonary fibrosis, whereas ones in control group were treated with same volume of normal saline. Each group was given corresponding medicines in the following day upon the completion of the model. Six rats in each groups were sacrificed on the 7th day and 28th day and sampled bronchoalveolar lavage fluid. Then the pulmonary tissue was removed, and HE staining and Masson staining were performed to observe pathological change of pulmonary alveolitis and pulmonary fibrosis. The extents of HMGB1, IL-8, a-SMA in bronchoalveolar lavage fluid were measured by enzyme-linked immunoadsordent assay. Experimental data related with SPSS 19.0 statistical software for statistical processing.Results:The Szapiel score among model group and the medicated groups showed increased compare with sham operation group, and that of the model group was the most obvious (p<0.01); Compare with model group, medicated groups’s pulmonary alveolitis and pulmonary fibrosis have different improvements with time(p<0.01 or p<.0.05); Pulmonary alveolitis score of Buyanghuanwu decoction group were higher than that of prednisone group on the 7th day (p<0.05), but had a lower on the 28th day (p<0.05), and so also were Pulmonary fibrosisat score of Buyanghuanwu decoction at same time point (p<0.05). Compared with the sham operation group, the expressions of HMGB1 in bronchoalveolar lavage fluid of model group were obviously increased (p<0.01). Each medicated groups had significantly decreased by comparison with model group (p<0.01). Compared with the prednisone group, the expressions of HMGB1 of Buyanghuanwu decoction group were higher than that of prednisone group on the 7th day(p<0.05),but had a lower on the 28th day (p<0.05). Compared with the sham operation group, the expressions of IL-8 in bronchoalveolar lavage fluid of model group were obviously increased (p<0.01), which on the 28th day were lower than on the 7th day, but were higher than the same period in the sham operation group yet. Each medicated groups had significantly decreased by comparison with model group (p<0.01 or p<0.05). Compared with the prednisone group, the expressions of IL-8 of Buyanghuanwu decoction group were higher than that of prednisone group on the 7th day and 28th day (p<0.05). Compared with the sham operation group, the expressions of α-SMA in bronchoalveolar lavage fluid of model group were obviously increased(p<0.01), which on the 28th day were higher than on the 7th day.Each medicated groups had significantly decreased by comparison with model group (p<0.01). There are no significantly differences with the expressions of a-SMA between prednisone group and Buyanghuanwu decoction group (p>0.05). But Buyanghuanwu decoction group had a lower than prednisone group on the 28th day(p<0.05).Conclusion:1. Intratracheal instillation of BLM can successfully construct mouse model of pulmonary fibrosis.2. Buyanghuanwu decoction could through suppress the excess expressions of HMGBl,IL-8,a-SMA effectively to alleviate alveolar catarrh and pulmonary fibrosis. That was proved Buyanghuanwu decoction had effect to anti-pulmonary fibrosis.
