ObjectiveTo investigate the relationship of serum 25-hydroxyvitamin D[25-?OH?D]level and gene polymorphism of vitamin D receptor?VDR?Fok? with Henoch Schonlein purpura?HSP?in children.MethodTo collect HSP patients in the Children's Medical Center,Affiliated Hospital of Qingdao University from Feb.1st,2017 to Dec.31st,2017.Totally 107 cases were included,who were randomized into none renal damage group?NHSPN,79 cases?and renal damage group?HSPN,28 cases?;and 76 health children were selected as the control group.Theserumlevelof25-hydroxyvitaminDwasdetectedby electrochemiluminescence,while the single nucleotide polymorphism of Fok? was detected by Sanger sequencing.Consequenceas1.The serum levels of 25-?OH?D were significantly decreased in HSP group comparing with the control group[t=-19.90,P<0.05];The serum levels of 25-?OH?D were significantly decreased in HSPN group comparing with the NHSPN group[t=3.35,P<0.05];no significant difference was noticed in serum 25-?OH?D level between the HSP group and control group[t=1.88,P>0.05;t=0.05,P>0.05].2.There are three genotypes of the VDR gene sequence,which are the wild type?TT?,TC mutant type and CC mutant type.The distribution of the three genotypes showed no difference in the HSP and the control group?X2=0.76,P>0.05?;no difference of allele frequencies of T and C was noticed in the two groups?X2=0.63,P>0.05?.3.There was no significant difference in the distribution frequencies of TT,TC and CC genotype at T\C locus of VDR FokI between NHSPN group?15.19%,60.75%,20.06%?and HSPN group?17.86%,60.71%,21.43%??X2=0.15,P>0.05?;no difference of allele frequencies of T and C was noticed in the two groups?X2=0.12,P>0.05?.Conclusion1.The decreased level of serum 25-hydroxyvitamin D may be related with HSP group and HSPN group;2.VDR Fok? gene locus TT,CT,CC genotype and T\C allele polymorphism may not be related to the pathogenesis of Henoch Schonlein purpura and purpura nephritis.
